Compared with that in Escherichia coli,the phosphoenolpyruvatesugar phosphate transferase system(PTS)in Gluconobacter oxydans contains the common components,El,HPr and EIIA,but lacks EIIB and EIIC functional groups.We plan to explore the function of PTS components in G.oxydans by enhancing or repressing their genes expressionRecently,small non-coding RNAs have already beenshown to typically either repress or activate gene expression in Escherichia coli.Here,artificially engineeredsmall RNAs to modulate gene expression in Gluconobacter oxydans were constructed.Firstly,three regulatory systems which can repress or activate the reporter gene expression were constructed when GFP was used as a reporter protein.Then we selected the component El protein of PTS in G.oxydans DSM2003 as the target protein.Based on the regulatory systems,we further designed a series of specific sRNAs and screened two sRNAs which could repress El expression effectively.It was the first report about the gene regulation by sRNA in G.oxydans.
