Agastache rugosus is a perennial herb of the family Lamiaceae.As a traditional Chinese medicine,its medicinal efficacy is mainly to relieve heat and treat diseases such as cold and heat.this experiment aims to separate good components from Agastache rugosus.Analysis of biological activity and chemical composition of active endophytic fungi and their secondary metabolites.In this experiment,rutin was used as a research object to study the isolation and purification of endophytic fungi,antioxidation and antibacterial activity of secondary metabolites,molecular biological identification and chemical composition of secondary metabolites.The research contents and results are as follows:Eight strains of endophytic fungi were isolated from the stem of Agastache rugosus,a medicinal plant,using a tissue separation method.seven strains were identified based on morphology.They were Aureobasidium sp.Alternaria sp.Stemphylium sp.Colletotrichum sp.and Corynespora sp.The eight isolated endophytic fungi were fermented and filtered to obtain mycelia and fermentation broth.The mycelia were extracted under reduced pressure and evaporated to gain ethanol extraction sites.The ethyl acetate and n-butanol were used to extract the fermentation broth,respectively.,Rotary vortexing to obtain broth ethyl acetate and n-butanol extract.Determination of the biological activity of secondary metabolites Three antioxidative activity experiments and in vitro bacteriostatic activity were studied using microbroth culture methods.The antioxidant results showed that most of the ethyl acetate fractions had antioxidant activity.The DPPH free radical scavenging results showed that the HX-J7 had a DPPH clearance rate of 97.44%,which was comparable to that of Vc.The total reducing power test results showed that the Vc equivalent exceeded.30 ?g/mg or more of the ethyl acetate fractions of HX-J1,HX-J4,HX-J6,HX-J7 and HX-J7 had a Vc equivalent of 52 ?g/mg;the results of hydroxyl radical scavenging experiments showed that HX-J2 The scavenging activity of ethyl acetate at HX-J6 and HX-J7 exceeded 50%,and the HX-J7 clearance was 58.30%.The n-butanol sites of HX-J6 and HX-J7 have a HX-J7 clearance rate of 72.88%.The results of antibacterial activity showed that the MIC values of HX-J7 for the six tested strains were 250,250,500,250,250 and 250 ?g/m L,respectively.In this paper,the ethyl acetate extract from the Agastache rugosus endophytic fungus HX-J7 fermentation broth was taken as the research object,and its chemical constituents were isolated and identified.Two compounds were isolated using silica gel column chromatography and Sephadex LH-20 chromatographic methods combined with thin layer chromatography.Using NMR,MS and other modern spectroscopy techniques,combining its physicochemical properties to analyze its structure,identified two compounds,namely: p-hydroxybenzoic acid(1),Ergosta-4,6,8(14),22-tetraen-3-one(2).Due to time,other parts are completed in the follow-up work.
