Blueberry anthocyanin is a kind of natural pigment, with better anti-tumor effect,but its physical and chemical properties are unstable, easy to changes under someexternal factors. Folate receptor can bind to the tumor cell surface, so it is often usedas a carrier of anticancer drugs targeting ligand. Therefore, to prepara a kind ofnanospheres with blueberry anthocyanins with the navigation folate, which canaddresses the blueberry anthocyanins poor stability issues, but also developed a newtargeted anti-cancer compound, to use the blueberry anthocyanins instead of thetraditional targeting anti-tumor carrier to make other complex vector.The FA-CS-ANNP is non-toxic, high efficacy, so can be used in the field of cancer treatment.Themain contents of the research are as follows: prepare a chitosan conjugates of folicacid as a targeted anti-tumor function of wall material, and use response surface tooptimize the preparation, use the spectrum to characterise the product.Using the folic acid-chitosan as a carrier conjugates to prepared nanosphereswith blueberry anthocyanins and optimizing the process parameters; while calculatingthe embedding rate and drug loading; by SEM, TEM, infrared spectroscopy,and laserparticle size analyzer to study the product characterization.By studying on the different temperature, pH and the rate of release ofanthocyanin to study the stability of the microspheres; examine the the case ofsustained release of the nanospheres in the artificial gastric juice. Studies have shownthat temperature, pH effects on the stability of the microspheres, and the microsphereshave a good release effect in the simulated gastric juice.Study on the function of microspheres: using the MTT assay to study on thefunction of anti-tumor of the nanospheres, the cells model are cervical cancer cell(Hela)?gastric cancer (SGC7901) cells and colon (Lovo); establish S180tumor-bearing mouse model to study on the distribution of thefolate-chitosan-anthocyanins(FA-CS-AN) microspheres in mice; FA-CS-ANnanospheres labeled with fluorescein isothiocyanate (FITC) were co-cultured withHela cells?and then survey with a microscope. The results show that nanoparticleshave the function of targeting anti-tumor.
