Human milk fat(HMF) has a unique composition to provide newborn infants with essential nutrients. Nevertheless, in cases when breastfeeding is not enough available, commercial infant formulas are the best alternative to fulfill the nutritional needs of newborns. To achieve high quality of HMF substitutes in infant formulas, there is great interest in the synthesis of 1,3-dioleoyl-2-palmitoylglycerol(OPO) recently. Until now, Israel AAK and Netherland Unilever are only two companies to provide OPO products. The Chinese Ministry of Health allowed OPO-type HMFs to add to infant formula.Alternatively, inexpensive compound cooking fat(CCF) is able to be used as a substrate to obtain OPO because the main chemical structure of CCF is similar to that of OPO. Thus, great investigations have been focused on the preparation of market demand of triacylglycerols(TAGs) enhanced with OPO from CCF.The goals of this study were to increase the abundance of OPO in TAGs and to establish a reliable method for characterization of OPO.Transesterification of CCF catalyzed by potassium palmitate gave PPP-enriched TAGs with a sn-2 positional palmitic acid content of 65% in the product. PPP-enriched TAGs were used as substrate for acidolysis catalyzed by Lipozyme RM IM. The OPO-type HMFs were successfully producedunder optimal conditions: a substrate molar ratio of 1:8(oleic acids to TAGs),and 22 h of reaction time at 60?. Under the optimal parameters mentioned above, the content of OPO was up to 43.8% in the final product.
