Corn bran is one of the largest byproducts in corn starch processing, and is mostly used as feed or fertilizer,so its economic value was lowered. Corn bran was hydrolyzed to be functional monosaccharide(L-arabinose, D-xylose, D-glucose), which can improve utilization of corn bran, providing a theoretical basis for recycling of corn processing by-products and a new idea for the development of functional monosaccharide. This paper studied the hydrolysis process of corn bran, separation and purification treatment of sugar liquid obtained by the optimal process.Using calcium resin separated the purificatory monosaccharides.1.In the process of ultrasonic-microwave synergistic hydrolysis of corn bran, the center combine experimental design was used to provide experimental data and analyzed to obtain the best technology parameter. The results showed that the reducing sugar content was 65.82% when the acid concentration was 3%, extraction time 60 min, microwave power 545 W and material-to-liquid ratio of 1:30(g/m L),which is increased 23.74% than that without ultrasonicmicrowave treatment. The analysis of acid hydrolysates by ion chromatography indicated that the content of arabinose was 20.74%, glucose was 11.73%, and xylose was 33.77%. FTIR analysis of the solid remains after corn bran degradation displays that there is not seriously destruction of the molecular structure of cellulose and found redshift phenomenon under four different processing. XRD indicates the ultrasonic-microwave synergistic treatment is the strongest degradation and crystallinity is minimum.13C-NMR and SEM show that the ultrasonic microwave synergistic treatment is superior to conventional degradation process, therefore, more reducing sugar is obtained.2.In the process of acid hydrolysis of steam explosion treated corn bran, the optimum parameters of steam explosion were obtained by orthogonal experiment that steam explosion pressure 1.0MPa, pressure time 20 s, material moisture 16.39%, and the reducing sugar content is 42.98%. Selected the optimal parameters to treat corn bran and then hydrolyzed, by the Box-Benhnken center combine experiment, the optimal degradation condition were received as follows: sulphuric acid concentration 1.66%, extraction time 1.5h, temperature 120? and material-to-liquid ratio of 1:10(g/m L), the yield of reducing sugars is 54.68% higher than 9.58%without steam explosion treated. Analysis of acid hydrolysates by ion chromatography indicatedthat the content of arabinose was 11.65%, glucose was 21.73%, and xylose was 17.99%. FTIR analysis of the solid remains after corn bran treated by steam explosion degradation displays that there is not seriously destruction of the molecular structure of cellulose and found redshift phenomenon under steam explosion. XRD indicates the steam explosion treatment is the strongest degradation and crystallinity is minimum. 13C-NMR and SEM show that the steam explosion treatment is superior to conventional degradation process, however, less than the ultrasonic-microwave synergistic treatment.3.Using activated carbon as decoloring agent, through the single factor experiment obtained the maximum decolorization rate of hydrolysate solution, the results show that activated carbon dosage was 20%, temperature 60?, decolorization time 40 min, oscillation frequency180 r / min. The maximum decolorization rate was 98.86% and loss rate of reducing sugar was20.43%.4.Desalting experiment has been selected D301 macroporous weakly basic anion exchange resin and 001*7 strongly acidic styrene type cation exchange resin. After four times cation exchange and twice anion exchange, the degradation solution can be purified up to the standard, and the conductivity is 92 ?s/cm.5.The Dow 99Ca2+ resin as filler, through the single-factor experiments, the optimal separation condition were received as follows: the column temperature 60?, sample flow0.4m L/min, 40% of the sample concentration and sample volume 5 m L. The maximum resolution is 0.46 between glucose and xylose, and 0.61 between xylose and arabinose.
