With the increasingly serious energy crisis and global food security issues, the fermentation manufacture of succinic acid by fiber hydrolyzate as raw materials is gradually becoming a hot research due to its advantages like low energy consumption, environmental friendly and sustainable use of raw materials. At present, research about fermentation production of succinic acid from fiber hydrolyzate as raw materials is still relatively scarce, especially the aspects of optimization of fermentation conditions. Hence, this experiment tested corn stover hydrolyzate fermentation of succinic acid conditions, combined with a response surface methodology, to improve the production of succinic acid ultimately. By this experiment, we not only improved the fermentation conditions of fermentation manufacture of succinic acid from fiber hydrolyzate as raw materials at some extent, but also provide references and technical support to realize industrialization of the fermentation production of succinic acid based on the raw materials of fiber hydrolyzate.By contrast three different facultative anaerobes, and obtained in three different facultative anaerobes, Anaerobiospirillum succinogenes fermentation is the best results,40.18 g/L(glucose concentration in the hydrolysis of the sugar concentration is 17.83 g/L,xylose concentration of 22.25 g/L) of corn stover hydrolyzate up to 7.86g/L,the maximum yield of the raw material fermentation of succinic acid and cell growth effect best, succinic acid conversion rate and succinic acidgeneration rate is the highest. Therefore, it obtained by comparing the Anaerobiospirillum succinogenes has a certain potential for industrial fermentation, and suitable for further research and application. In the process of optimization of culture conditions further on the Anaerobiospirillum succinogenes fermentation succinic acid obtained hydrolyzate concentration of 39.99 g/L of corn stover hydrolyzate as raw materials, put on the Anaerobiospirillum succinogenes fermentation succinic acid fermentation process conditions optimization, draw the liquid volume in the medium in a succinic acid fermentation process is the 300 ml anaerobic bottled mounted 90 ml, succinic acid production for up to 8.5 g/L; when succinic acid fermentation culture temperature is 37℃, the highest bacterial OD660, succinic acid production has reached up to 8.42 g/L; when the initial p H of 6.8, the highest yield is 8.75 g/L, and the cell growth is also good; when the Anaerobiospirillum succinogenes fermentation inoculation amount was 11%,succinic acid production to achieve highest 13.37 g/L,OD660 is relatively high.On the basis of the best fermentation conditions,and using response surface methodology to optimize hydrolysis sugar mass concentration,corn syrup mass concentration and Na2CO3 added in the fermentation medium. After regression analysis of variance and regression analysis, the optimal conditions: the hydrolyzed sugar 60 g/L, corn steep liquor 10g/L, Na2CO3(concentration for 10 mol/L) addition amount of 2ml, the predictive value of 19.6134 g/L. After fitting the validation of the test,he production of succinic acid up to 18.32 g/L. Verify that the value and optimize the value of the response surface model is basically the same.Optimization model for better reliability. Finally, the optimum fermentation and nutritional conditions, corn stover hydrolyzate of hydrolysis sugar concentration of 59.66 g/L as raw material, the use of Anaerobiospirillum succinogenes batch fermentation of succinic acid in 1000 ml anaerobic bottled,the final fermentation maximum yield of 19.66 g/L, Batch fermentation in the 1000 ml anaerobic bottled amplification test to obtain a higher concentration of succinic acid, and improve the Anaerobiospirillum succinogenes fermentation production.In this study, the main advantage of the research is rely on the sustainable use of raw materials and cheap industrial nitrogen source, and if it could realize industrialization of the fermentation production of succinic acid. It would effectively reduce the dependence on starch raw materials to fermentation manufacture of succinic acid. The application of the research would not only get rid of dependence on fossil fuels, but also develop new ways to use of the greenhouse gas carbon dioxide, make succinate become the most important future bio-based chemical products.
