As an crucial tumor suppressor gene, BRCA2 has a close relationship with many kinds of cancer and plays an important role in the maintenance of genome stability. Those eight repeat motifs(BRC) of BRCA2 have the ability to interact with the RAD51 protein, which may affect the repair of damaged single stranded DNA. So the importance of researching the interaction of BRC and RAD51 is very obvious.In this paper,we obtained the posssile binding mode of BRC8 and RAD51 based on the possible structure of BRC8 utlizing the molecular docking method named ZDOCK and found those potienl active sites of polypeptide through the vitual alanine screening,then fourteen polypeptide were designed. We also purified and identified the structure of nine polypeptids including BRC8 and target peptide and BRC8-like peptides with HPLC and ESI-MS.At last, by using CD, we found some peptides had a good performance on interacting with the targed peptide. By using circular dichroism analysis software named CDPro analyze the target peptide and peptide-like interaction after the target peptide secondary structure, We found that ?-helix content of the peptide BRC8-2 and BRC8-3 and BRC8-6 with target peptide increased by 23.6%,15.1%,13.5%. Therefore we consider that those peptides have a stronger role in interaction with target peptide.
