Obesity,closely related to the various metabolic diseases which serious threat to human health,is fat accumulation in the body caused by intake of food energy more than consumption,such as hypertension,type ?diabetes,heart disease and some cancers,so seeking safety effective foods that can reduce weight is one of hot fields for sciencfic worker in foods.The recent research showed that Pu'er tea has a certain effect of loss weight and fat;however,exact ingredients of reducing weight of Pu'er and its mechanism are unclear.Since,it is the very vital significance for studying Pu'er pigment effects on reducing weight and acting mechanism.Pu'er pigment effects on reducing weight and lipids were researched by detecting cholesterol(TC),triglyceride(TG),High-density single sterols(HDL-C),Low density single sterols(LDL-C)in the serum and body weight,body fat weight,lee's index of the rats with nutritional obesity model of Sprague-Dawley rats which treated with different concentrations of tea pigment for six weeks.The results showed that tea pigment could decrease body weight,body fat weight,Lee's index,the rat weights decreased 30.35% in contrast to the control,while treat with high-dose pigment.The concentrations of TC,TG and LDL-C were all decreased significantly,the serum level of HDL-C increased remarkably after treated with tea pigment,the high concentrations of cholesterol blood and hyperglycemia in bood that caused by obesity,droped to the proximity level of normal rats.The size of adipocyte and fat accumulation in the liver tissue of mouse which use drug interention were also observed by paraffin,the results indicated that tea pigment can reduce the size of adipocyte and reduce fat accumulation in the liver tissue.Hence,tea pigment could have some preventive and curative functions of hepatopathy arising from obesity.Pancreatic lipase is a key enzyme in the digestion and absorption of lipids of food,the influence of tea pigment on pancreatic lipase activity can be researched for illuminating its acting mechanism of reducing weight,by detecting the color changes of pancreatic lipase reaction before and after,its substrates was the nature fat analogues 2,4-dinitrophenyl butyrate(DNPB).The results showed that the activity of pancreatic lipase was inhibited notablely,while the tea pigment concentration range0.02-0.1(mg/ml).And more than 52% the activity of pancreatic lipase could be inhibited,when tea pigment concentration than 0.06 mg/ml,reaction time 5 min.The levels of adipocytokines were determined by using liquid chip technology and ELISA method,such as IL-1?,IL-1?,IL-6,Leptin,TNF-?,VEGF,Adiponectin,Insulin,for explaning the mechnisms of losing weight of Pu'er pigment.Results showed that tea pigment could reduce the concentrations of IL-1?,IL-6,Leptin and TNF-?,Insulin,VEGF,and improve those of IL-1?,adiponectin.Therefore,adjustment the levels of adipocytokines may be the one mechanism of lose weight of tea pigment.
