Trans fatty acids produced by the processes of food processing have potential threat to human health.Previous research has shown that dietary intake of trans fatty acids could cause obesity.In the early development of adipose tissue,the intake of trans fatty acids was positively related to the morbidity of adult obesity.The adipogenic differentiation of adipose tissue-derived stromal cells(ADSCs)plays a vital role in the development of adipose tissue.However,the effect and its mechanism of trans fatty acid on adipogenic differentiation in ADSCs were still unknown.In this study,we used linoelaidic acid,as well as linoleic acid and linolenic acid,to evaluate the effect of trans fatty acid on adipogenic differentiation in ADSCs,its mechanism also were determined.This study was to provide scientific basis for further study of the relationship between the intake of trans fatty acids and the occurrence of obesity or other abnormal lipid metabolism.ADSCs were isolated and cultured in vitro,and then induced into adipocyte.Oil red O staining and TG connect assay were used to analyse the effect of linoelaidic acid on adipogenic differentiation in ADSCs.Our results have shown that linoelaidic acid could incerase the adipogenic differentiation in a dose-dependent manner in the concentrations of 20~60 ?mol/L,compared with linoleic acid and linolenic acid.Furthermore,we found that linoelaidic acid significantly downregulated the expression of ?-catenin and inhibited the accumulation of ?-catenin proteins in nucleus by Western blot and immunofluorescence assay,respectively.These results suggested that the stimulative effect of trans fatty acids on adipogenesis in ADSCs may be involved in Wnt/?-catenin signaling pathway.Li~+ was used as an activator of Wnt/?-catenin pathway.Combined with oil red O staining,TG connect assay,Western blot and immunofluorescence assay,we found that supplement of Li~+ could antagonize the effect of linoelaidic acid on adipogenesis in ADSCs,up-regulated the expression of ?-catenin and increased the accumulation of ?-catenin proteins in nucleus.In conclusion,our results indicated that linoelaidic acid could enhance the adipogenic differentiation in ADSCs in vitro,which is partly due to the downregulation of ?-catenin and suppression of Wnt/?-catenin pathway.
