With the increasing of population and human activities,our destruction of the marine environment has become increasingly intensified,all kinds of pollutants cause serious problems in the sea,and these problems begin to restrict the sustainable development of the marine economy and society.Biological monitoring is produced by the community and population or individual biological response to environmental changes,a monitoring method with victory changes to reflect the pollution condition.The application of biomarkers in environmental monitoring,can obtain the mutual information about chemical exposure,biological effects,the effects of pollution,early warning and assessment of the environment.Chlorella and Dunaliella salina is a species of phytoplankton and algae cells containing protein,polysaccharide,lipids and many kinds of inorganic elements,cell metabolism is fast,the application value is high.In this paper,the sea water in the surrounding area of Dalian is studied as the research object,using Chlorella and Dunaliella as experimental materials,UV spectrophotometer method for the determination of microalgae growth curve and chlorophyll content,protein content was determined using Coomassie brilliant blue staining in algae cells,determination of polysaccharide content in microalgae using anthrone colorimetric method,then the relationship between the water sample and the physiological biochemical indexes of microalgae is analyzed by the method of grey correlation analysis.The research shows the results as follows:?1?Measure and analysis the physical and chemical of seawater samples collected from 8sampling sites in the sea area of Dalian,Liaoning Province.It is concluded that the salinity difference between the 8 sampling points is not large.The BOD5 content of the overall high in addition to Xinghai Square.The low dissolved oxygen content of the Donggang business district,Xinghai Square and tower bay beach area.The highest oil content was found in fourth sampling point.That is because the waters near the tower bay beach belongs to inshore fishing zone.There is a large number of fishing boats in the long term give rise to the high oil content in the sea area.The contents of nitrate and nitrite is high in Xinghai Square.The petroleum content,anionic surfactant content of Lushunkou District Gold Hill Beach is high.The content of total phosphorus,total nitrogen,petroleum,nitrate and nitrite of Lushunkou District Long River Estuary was high.Because the sea area belongs to the river to the sea,sothe organic pollution levels in this sampling point is higher than other sampling points discharged into the sea by rivers.?2?Cultivation of Chlorella and Dunaliella salina with 8 sampling points of seawater samples,The cell number and biochemical parameters were determined,The data were analyzed that: the growth state and biochemical index state of Chlorella have changed significantly with changes in the external environment.The water samples collected in Donggang business district,Xinghai Square,Lushunkou District Gold Hill Beach inhibition of protein content in Chlorella growth.The number of cells and the biochemical indexes of the cells did not change significantly with the changes of seawater samples.This shows that Chlorella is more suitable for biomarkers of seawater.The pollution status of the waters can be evaluated according to the determination of the number and biochemical indexes of the Chlorella.?3?Through the DPS data processing system,correlation analysis between biochemical indexes of microalgae and the biochemical indexes of seawater.Results show that: total phosphorus,total nitrogen and nitrite were closely related to the number of microalgae cells.There is a high correlation degree between anionic surfactant and micro algae chlorophyll a.In the early stage of microalgae culture,petroleum,nitrite,total phosphorus and total nitrogen have a high degree of correlation the change of Chlorella polysaccharide.In the early metaphase of microalgae culture,the correlation of N and P elements in seawater with the content of protein in microalgae cells was high.In the late stage of microalgae culture,the salinity of sea water and the index of anionic surfactant have high correlation with the protein content of microalgae.The concentration of total phosphorus,total nitrogen,petroleum and nitrite with the concentration of polysaccharide was decreased.
