Cardiovascular and cerebrovascular disease are serious threat to human health.Currently,every year about 18 million people from all over the world die from cardiovascular and cerebrovascular diseases.Western medicine has many side effects,but traditional Chinese herbal medicine is loved by patients for its less toxic advantage.Polyphenols are important components of Chinese herbal medicine.Their therapeutic mechanisms of cardiovascular and cerebrovascular diseases are focus on by some researchers.And their therapeutic mechanisms are mostly attributed to polyphenols' antioxidant activity former scientists.Recently,excess endogenous aldehydes'(especially formaldehyde endogenous)accumulation in the body is believed to another risk factor during the process of cardiovascular and cerebrovascular diseases.This study uses gallic acid as the representative of natural polyphenols,studying the adducts of it scavenge formaldehyde,then confirming gallic acid can reacte with formaldehyde in vitro,to acheive prevention and treatment of cardiovascular and cerebrovascular diseases.This will be specific proof of polyphenols mechanism for prevention and treatment of cardiovascular and cerebrovascular diseases.It also can provide new ideas for natural polyphenol metabolism and pharmacodynamics.In this study,gallic acid and formaldehyde were reacted with the amino acids(phenylalanine,glutamic acid,lysine)of different acid-base properties,under relatively mild conditions,researching the product to uses HPLC-MS,NMR.Finally,the four new Mannich base derivatives: MC(1),MC(2),MC(3),MC(4)were obtained.Gallic acid-formaldehyde-amino acid monomeric adducts were found [M-182]+ fragmentation characteristics from their mass rule.And gallic acid and formaldehyde are added into fetal calf serum for reaction which was detected by liquid chromatography-mass spectrometry.Eventually,the polyphenols(gallic acid)-formaldehyde-amines(amino acid)three-component addition products.were found in fetal bovine serum reaction system.
