In this experiment,two varieties of Perilla planted by our own(Shijiazhuang and Liaoning)were used as experimental materials.Four kinds of main active components(perilla ketone,rosmarinic acid,lutein,beta carotene)of different parts and different harvested months were determined,compared with the medicinal perilla leaf and stem purchased from pharmacy at the same time.We alse evaluated the antioxidant activity of the crude extracts from perilla leaves.The results were as follows:By the headspace solid-phase microextraction and gas chromatograph-mass spectrometer(GC/MS),we confirmed the chemical type of the two varieties of Perilla--Shijiazhuang and Liaoning varieties.The two varieties of Perilla used in this experiment belong to the PE type.We separated and purified the perilla ketone by high speed countercurrent chromatography,the purity of the perilla ketone was 99.711%.We confirmed the optimum extraction conditions of rosmarinic acid,lutein and beta carotene in Perilla.The main active ingredients(perilla ketone,rosmarinic acid,lutein and beta carotene)in Perzilla of different harvested monthes and different harvested sites(the leaves,stems and seeds)were determined,compared with the medicinal Perilla leaf and stem purchased from pharmacy at the same time.The results showed that four active ingredients were both mainly distributed in Perilla leaves.Comparison of the active ingredients in different harvest periods,the content of active components in the samples collected at the end of July was relatively high.The contents of four active components in the leaves and stems of the pharmacy were in the middle,compared with the samples of different harvested periods.We studied the content variation of the active ingredients in Perilla leaf after treated with different pretreatment methods of raw materials(freeze-dried,40? drying,60? drying,80? drying and 100? drying).Comprehensive consideration of economic benefit and protection of the active components,the drying temperature of Perilla leaves is proposed no more than 40 ? during processing.We extracted rosmarinic acid from Perilla leaves of Shijiazhuang,the content of rosmarinic acid was 92.941 mg/g in Perilla leaves extract.The antioxidant capacity of Perilla leaf extract and pure rosmarinic acid was evaluated with HepG2 cell.The results showed that Perilla leaf extract has stronger antioxidant capacity.It is suggested that besides rosmarinic acid,there are other active ingredients playing a significant role against oxidative effect in Perilla leaf extract.
