Preserved egg,as one of our traditional pickled eggs,according to the record,have an effect of falling blood pressure,at present,the research of extracting directly from the vinegar egg and using enzymatic hydrolysis eggs or protein powder to prepare angiotensin enzyme(ACE)inhibitory peptides is more,but the study of ACE inhibitory peptides in the preserved eggs is less.In this study,decompression preserved eggs(DPE)for raw materials,compared with the normal pressure method preserved eggs,the basic indicators and protein degradation was analyzed.Using direct extraction method,preserved egg white high activity of angiotensin enzyme(ACE)inhibitory peptide was extracted,gel chromatography and mass spectrometry identified five ACE inhibitory peptide,and the peptide extraction gavaged essential hypertension(SHR)rats,which showed a hypotensive activity.The preparation of decompression preserved egg:The orthogonal analysis method used egg white,egg yolk nonprotein nitrogen content as the indexes,the optimal combination is A3B3C2.NaOH content of 4.2%,the salt content of 5.5%,copper content of 0.40%,is the optimum conditions of processing eggs.In the decompression and atmospheric preserved process,DPE is slightly higher than the total amount of atmospheric moisture,pH value in egg whites was similar,the yolk pH value in DPN is slightly higher than DPE,the overall trend of the total salt content is consistent,which showed a gradual increasing trend,free amino acids in DPE is slightly higher than DPN's.Egg total nitrogen content decreased,non-protein nitrogen,amino nitrogen,nitrogen and polypeptide TVB-N increased gradually.Until after 60 days of decompression and preserved eggs,total nitrogen content compared with fresh duck eggs decreased by 7.03%and 13.51%,non-protein nitrogen content increased by 90.91%and 137.71%.The separation of preserved egg ACE inhibitory peptides:Single factor test cleared that the extraction time was 240 min,extraction temperature was 50?,extraction solid-liquid ratio 30mL·g-1 and extraction pH of 8.0.Take 3000Da less coarse filter material,component b was isolated and purified by Sephadex G-15,subjected to mass spectrometry,and identified 3 peptides from the ovalbumin(LINSWVESQTNGIIRN,SVSEEFRADHPFL and SVSEEFRADHPFLF)and 2 peptide from the ovotransferrin(NKVEDIWSF and LFGPPG).The vivo tests of ACE inhibitory peptides in preserved egg:Spontaneously Hypertensive Rats fed egg ACE inhibitory peptides six weeks,captopril positive control group and low,medium and high dose groups at each time blood pressure remained stable,slow growth,there was a significant statistical difference(P<0.05)compared with the control group,blood pressure increased by 3.70 mmHg,15.66mmHg,3.33mmHg,5.60mmHg,the control group increased by 19.80 mmHg.Eggs ACE inhibitory peptides significantly reduce total cholesterol levels,increased high-density lipoprotein cholesterol levels,and makes angiotensin-converting enzyme and endothelin-1 showed a significant reduction(high dose group than in the control group decreased 110.99 and 27.91 ng/mL).SHR rats' liver was determined:total 10 kinds of saturated fatty acids,five kinds of monounsaturated fatty acids and four kinds of polyunsaturated fatty acids,as compared with the control group,while the low,medium and high dose groups of unsaturated fatty acids in the fatty acid the proportion of the total substantially gradual upward trend.
