Nanoparticles can be used to analyze the trace substances of living systems,can be obtained the biochemical information at the single cell level and the single-molecule level,that have a great importance for understanding biomolecular structure,function,and clarifing the physiological and pathological of disease and even early diagnosis of the disease.Inductively coupled plasma mass spectrometry(ICP-MS)have a lot of advantages,such as high sensitivity,accuracy and simultaneous multi-element analysis.ICP-MS plays a huge role in the detection of trace and ultra-trace elements,it also has great potential in the biochemical analysis.In this paper,we used the ICP-MS to make a elemental imaging analysis for biological tissue samples;synthesized nanoparticles label and established a competitive immunoassay method based on the ICP-MS to measured ferritin levels in Serum.That will lay the foundation for the quantitative image analysis of biological in the future.This approach will have broad applications in the rapid diagnosis of the disease,proteomics,genomics and other metals in the neighborhood.The main work is as follows:(1)LA-ICP-MS with a good spatial resolution,good detection analysis,operating cost advantages,have the ability to sample in situ analysis.This paper established the LA-ICP-MS imaging element method.The use of LA-ICP-MS for small Shushu brain slices Fe,Cu,Zn and other trace element distribution in rat brain were analyzed;of gold nanoparticles exposed to gold nanoparticles after the single cell within the natural elements Mg distribution of elemental imaging and optical imaging contrast unicellular want.LA-ICP-MS is expected as a strong complement to other biological imaging techniques to be more widely used in biomedical research,play a more important role.(2)An immunoassay is a biochemical test that measures the concentration of a biomolecule using an antibody-antigen reaction.Inductively coupled plasma mass spectrometry(ICP-MS)emerges as a promising detector for immunoassay.1 This ICP-MS-based immunoassay has great multiplexing potential because more than 100 isotopes are theoretically available for antibody labeling.2 One inherent limitation when using elemental labeling reagents is that only limited metal atoms can be attached to an antibody molecule,thus restricting immunoassay sensitivity.The use of nanoparticle as element labeling in ICP-MS analysis will increase signal intensity and provide more information with low levels of target proteins because each label can contain thousands of atoms.Here we developed a new competitive immunoassay of ferritins in this paper using lanthanide nanoparticle labeling and LA-ICP-MS analysis.
