The amide fungicides of isofetamid was reserrched and developed by a Japanese company in 2005.As a kind of SDHI fungicides,it plays a role which inhibiting its breath by effecting on the succinate dehydrogenase of pathogenic bacteria.The synthetic process of Isofetamid and its intermediates was introduced in detail based on literature review.Synthesis of intermediate 3-methyl thiophene-2-benzoyl chloride,?-amino-4-isopropyl-2-methyl isobutyl ketone benzene and target product Isofetamid,the intermediates and the target compounds were characterized.(1)The synthesis of 3-methylthiophene-2-carbonyl chloride which is 3-methylthiophene-2-formaldehyde as the reaction of the starting material,We obtained the reaction product of 3-methylthiophene-2-carboxylic acid which Oxidized by potassium permanganate,then it is Chlorinated with thionyl chloride to give 3-methylthiophene-2-carbonyl chloride.(2)In the synthesis of ?-amino-4-isopropoxy-2-methylbenzene isobutyl ketone,it uses m-cresol and isobutyryl chloride in anhydrous aluminum chloride as a catalyst through the action of grams of acylation to obtain 4-hydroxy-2-methyl benzene isobutyl ketone,followed by esterification with 2-bromopropane to give 4-isopropoxy-2-methylbenzene isobutyl ketone,and then with phenyl trimethyl ammonium bromide reaction of ?-bromo-4-isopropoxy-2-methyl benzene isobutyl ketone,The reaction was continued to give 2-[1-(4-isopropoxy-2-methylphenyl)-2-methyl]-1-oxopropan-1,3-dione with potassium phthalimide,it obtains ?-amino-4-isopropoxy-2-methyl benzene isobutyl ketone with hydrolysis under the conditions in the presence of hydrazine hydrate.(3)In the synthesis of Isofetamid,the two intermediates undergo an amidation reaction to give the desired product.Through feasibility studies on synthesis process of Isofetamid,we screened a more optimized process route,analyze the various specific factors affecting the product yield,obtained ideal synthetic route parameters,the entire synthetic route of the method is moderate,it has a very good development prospects for the production of products in the future which laying the foundation for the industrialization.
