The Research Of Shaddock Peel Polysaccharides On Tumor Inhibition

An orthogonal experiment was used to optimise the extraction conditions for polysaccharide from shaddock peel.The composition in polysaccharide from shaddock peel was determinated,and the structure of polysaccharide was elucidated by UV spectrum and IR spectroscopy.The monosaccharide composition of polysaccharide was determinated by gas chromatography.Animal models and cell models are established to investigate the anti-tumor activity of polysaccharide in vitro and in vivo.The optimum extraction conditions were as follows:water-to-raw material ratio,1:40,extraction time of 1.5 h,extraction temperature 95 ?.and extraction pH 2.The experimental polysaccharide yield was 17.6%.The crude polysaccharides contained 77.01%carbohydrates,and the polysaccharides was mainly composed of glucose.The result of MTT test showed that the polysaccharide almost have no inhibition on tumor cells in vitro.Using low dose(100 mg kg^-1 d^-1),middle dose?200 mg kg-d-1?and high dose(300 mg kg^-1 d^-1)to gavage the tumor-bearing mice,the result showed that the middle dose have the highest inhibition on tumor with the tumor inhibitory rate of 50%.Through blood index and immune organs index to study the antitumor effect of shaddock peel polysaccharide.The blood index including blood routine examination,peripheral blood lymphocyte subsets examination,serum antibodies detection and serum Cytokine detection.The immune organs index including observing the structure of immune organs,detecting the lymphocyte subsets of grinding fluid,carring on lymphocyte proliferation experiment and NK cell killing experiment.The macrophage phagocytic function on neutral red was also determined.Through above experiments,we can claime that:the shaddock peel polysaccharides can increase the tumor-bearing mice the proportion of lymphocyte subsets in blood,enhance the activity of lymphocytes,promote the specificity of antibodies and the secretion of cytokines,can well protect and maintain the form of immune organs in tumor-bearing mice,can promote the tumor-bearing mice lymphocyte proliferation,enhance the killing ability of NK cells.What's more,the polysaccharide can enhance macrophage phagocytic function on neutral red.Through the above experimental results,we can concluded that shaddock peel polysaccharides can not hurt tumor cells directly,its anti-tumor effect is through promoting the proliferation of lymphocytes,enhancing the activity of immune cells,promoting the secretion of antibodies and cytokine,protecting the immune organs and other ways to enhance the body's cellular immunity,humoral immunity and non-specific immunity.
