Pyrimidoquinolines as a very important heterocyclic compounds,widely present in the natural products and show good biological and pharmacological activity,these compounds are mainly used in pesticides,anti-bacterial anti-inflammatory drugs,and anti cancer drugs and so on.This paper describes that 2-aminoquinoline was considered as the object of study,which works on synthesis method of hetrocycle-fused quinoline derivatives,specific work is as follows:In the first part,the synthesis,activity and application of pyrimidoquinoline derivatives were reviewed on the basis of the survey of the relevant numbers of literatures.The second part,we used 2-aminoquinoline-3-carboxylate as raw material to synthesize pyrimido[4,5-b]quinoline-4(3H)-ones from condensation of 2-amino quinoline-3-carboxylate,N,N-dimethylformamide dimethylacetal,and primary amines without catalysts.The third part,a novel methodology for facile production of 2,3-dihydropyrimido[4,5-b]quinolin-4(1H)-ones by condensation between 2-aminoquinoline-3-carbonitrile with aldehydes in the presence of sodium hydroxide.The forth part,we describe an efficient one pot three-component protocol for the synthesis of pyrimido[4,5-b]quinolin-4-amines from 2-aminoquinoline-3-carbonitrile,N,N-dimethylformamide dimethylacetal,and primary amines under microwave irradiation.The structures of all compounds were characterized by elemental analysis,IR,1H-NMR spectrum and 13C-NMR spectrum.
