Moringa oleifera Lam.is a kind of special plant with market potential and development prospect in subtropical areas.It's leaves,flowers,seeds and tender shoots,tender stems and roots are all valuable edible and medicinal plants.Rich oil,protein and minerals are contained in moringa seeds and which can be used in products of food,medicine,cosmetic and lubricant.The refined products produced with moringa seeds are short of supply because of the lack of deep-developing of the oil and protein in moringa seeds.The extraction of moringa seed oil and protein hydrolysates completed at the same time with aqueous enzymatic method is talked in this article.Providing theoretical guidance for more valuable process of moringa seeds by studying demulsification process and slag washing process.Studying the effect of the stability of moringa seeds peptide influenced by the process condition based on producing moringa seed antioxidant peptides with aqueous enzymatic method.Taking the moringa seeds peptide as functional ingredient in the development of food to give theoretical and method guidance.(1)The technology conditions about improving the yield rate of moringa seed oil and protein hydrolysate with alkali pretreatment are confirmed in this article as follows: the best alkali rate pH is 8.0,alkali time is 60 min;taking the yield rate of free oil and the recovery rate of protein as index by adopting the single factor experiment,response surface experiment to optimize the technology of extracting moringa seed oil and antioxidant peptides simultaneously with aqueous enzymatic method.The total amount of enzyme is 26%,Ns37071 is 46.19%,enzymatic hydrolysis time is 24 H,the model estimated yield rate of free oil is 70.18%,the recovery rate of protein is 63.40%,DPPH eliminating ability is 132.99?mol Trolox equiv/100 g.Adjust the total amount of enzyme to 2.60%.Ns37071 is 4 6.00% in the total amount of enzyme.Enzymatic hydrolysis time is 24 H.The gained recovery rate of free oil is70.09±0.65%,the recovery rate of protein is 63.69±2.94%.DPPH eliminating ability is 139.38±0.96?mol Trolox equiv/100 g.(2)Adopting demulsification process to improve the yield rate of moringa seed oil with physical and chemical methods,choosing the way of heating demulsification to optimize the technology of extracting moringa seed oil and protein hydrolysate at the same time with the aqueous enzymatic method as follows: total amount of enzyme is 2.6%(Ns37071 is 46% in the total amount of enzyme),24 H enzymolysis with 55?,killing enzyme 30 min with 95?.After being cooled and 10000 g is centrifuged with 30 min.the yield rate of moringa seed oil reaches 76%.Studying the slag washing process by adopting the ultrasonic assisted extraction and alkali extraction to improve the protein recovery rate of moringa seed.Confirm the optimized slag washing process technology is 150 W ultrasonic 25 min,PH 10.0,3H extraction with 55?.The recovery rate of moringa seed protein can reach 72% combining the protein hydrolysate obtained by aqueous enzymatic method.(3)Studying the effect of the antioxidant activity of moringa seeds peptide caused by temperature,pH,metal ion,carbohydrate based on producing antioxidant peptides with aqueous enzymatic method.The experiment shows that if the moringa seeds peptide is heated with 115? for 20 min,its antioxidant activity can be improved obviously.The moringa seeds peptide's antioxidant activity is higher in acid environment.Under the acid environment,if the moringa seeds peptide is heated,its antioxidant activity can be improved higher.Under the non-heat treatment,as the rising of the metal ion's density,the antioxidant activity of moringa seeds peptide can be improved with different levels.With heat treatment,as the rising of the Ca2+?Mg2+?Zn2+?K+ the antioxidant activity of moringa seeds peptide can be lower a little bit.But the antioxidant activity of moringa seeds peptide can be improved obviously as the rising of Cu2+ density.The antioxidant activity of moringa seeds carbohydrate can be improved greatly by adding carbohydrate substances such as fructose,galactose,glucose,lactose,sucrose,mannose,maltose,GOS.The order of the carbohydrate substances that can help to improve the antioxidant activity of moringa seeds peptide is fructose>galactose>GOS>glucose>lactose=sucrose=mannose=maltose.
