Acarbose is a potent?-glucosidase inhibitor,It has been widely applied in treatment of type II diabetes.Recently,as the patients of diabetes increased sharply in our country,the demand for acarbose is growing quickly,which evident its promising market prospects.However,the production of acarbose by fermentation suffers the drawback of low productivity,high production costs.Therefore,improving the acarbose production through the industrial microbial fermentation and optimize the fermentation process,reduce production cost is the primary objective for current industrial.Through optimizing the Shake flask fermentation conditions by EastChina Medicine workshop107 production of acarbose strains as follows:medium loading 30ml per 250ml shaking flask,seed cultivation time 60 h,inoculum size of 10%?v/v?,initial pH 7.0,28?,200 rpm,under these conditions,the production of acarbose was raise to 4835.21mg/l.By the further optimized.The optimized medium?g/l?were as follow:glucose 30.0,saccharified liquid 312.5 ml?maltose 50.0?,soybean meal 15.0,sodium glutamate 6.0,CaCO3 2.0,CaCl2 3.0,FeCl31.2,K2HPO4 0.8.Under the optimum conditions,the production of acarbose was reached to 5250.40mg/l.Then,the 10m3 fermentation production of acarbose fermentation process were studied,we established an efficient fed-batch strategy,the initial fermentation medium was consist of?in g/l water?:glucose,10;soybean flour,17;maltose,60;glutamate,5;CaCO3,2.5;CaCl2,1.88;K2HPO4,1.0;FeCl3·6H2O,0.83;glycerol 5;fed 8g/l glucose respectively at 24 h,48 h,72 h,96 h,120 h,144 h,and fed 20 g/l maltose once at 96 h.At last,the acarbose titer reached to 5800 mg/l at 192 h.external addition of glutamate on the basis of the fed-batch can not only enhance the production of acarbose,but also have some inhibitions.Finally,we established the 60m3 fermentation production of acarbose feeding process.The continuous feeding compound nutrients is moreconducivetopromotethesynthesisofacarbose.Continuous-fermentation in 60m3 fermentor,the loading of 35m3,after fermentation 48h,began feeding at 3.5L/min of feed solution for 88h,then adjust the feeding amount down to 1.5 L/min,until 13.5m3,the acarbose yield reached 8139mg/L,then decreased with the prolonging of time.Component A is 43.1 mg/L,the relative content of acarbose was0.53%.
