Polypeptide is a class of biologically active macromolecules and intermediate of proteolysis,which is formed by amino acids through peptide bonds.Currently there are two ways to obtain the polypeptide,including biological gene transcoding and chemical synthesis.The chemical synthesis comprises solid phase synthesis methods and liquid phase synthesis method.Chemical synthesis methods mainly include solid-phase peptide synthesis,and long peptides or proteins are synthesised by polypeptides connection method.Polypeptide Connections method include native chemical ligation,hydrazide peptides connection method and auxiliary mediated peptide connection method.Polypeptide connection method in this paper is improved auxiliary mediated peptide ligation method combined hydrazide peptides connection method.Because of intramolecular cyclization,Asp,Asn and Gln can not be easily prepared.C-terminal other 16 peptide hydrazides were designed to react with glycine.N-terminal auxiliary group is a benzyl and amino acids involved in the reaction include glycine,alanine and isoleucine,glutamic and lysine.Experimental results show that auxiliary group mediated polypeptide ligation reaction is fit for amino acid of the low resistance,like glycine,not sterically hindered amino acid.And the linker reactions between different peptide hydrazine and glycine also showed different connection efficiencies.This paper systematically studied connections between peptide hydrazide and amino acids with auxiliary to provide a new connection idea for long peptide synthesis strategies.
