Curcumin,which was extracted from curcuma longa that is a medicinal plant widely cultivated in tropical regions of Asia has antioxidant,anti-inflammatory,anticancer,antirheumatic,antibacteral,antihepatotoxic,anti-Alzheimer s disease and anti-HIV activities.Efforts to improve the biological activities of curcumin have led to the development of curcumin analogues by appropriate structural modification of curcumin.In the work described here,we designed and synthesized two series of eighteen new compounds in order to introduce the indole group to N-substituted piperidone analogues of curcumin which was via Knoevenagel reactions.These compounds were characterized by UV~1H NMR,IR,and ESI-MS.The antioxidant activity of these compounds was investigated using the 1,1-diphenyl-2-picrylhydrazyl method.The results showed that the curcumin analogues exhibit varying degrees of antioxidant activity.Amongst the new compounds,C7,C8,C9,D8 and D9 showed higher free radical scavenger activity.And the anti-Alzheimer s disease activity of compounds D1 to D9 was investigated two different drug screening models for acetylcholinesterase inhibitors and butyrylcholinesterase inhibitors respectively.The results indicated that D3,D4 and D9 have higher anti-Alzheimer's disease activity.The crystal structure of C3 which was determined by single crystal X-ray crystallgraphy showed that the crystal is Triclinic and crystal parameters:a=7.7671(7)(?),b=9.1331(6)(?),c=12.4571(11)(?);?=84.541(7)0,?=83.814(7)°,?=66.482(8)0;Z=2;space group P-1;Final R indices Rl=0.0408,wR2=0.1134;R indices(all date)R1=0.0495,wR2=0.1210.
