Astaxanthin is one of the oxygenated compounds of carotenoid and also the highest level product of its compounding.It is a kind of ketonic type carotenoid,and is the most potential antioxidant pigment in nature.But due to the more conjugated double bonds in the weak polar molecule structure of astaxanthin,many external factors including lighting,oxygen and so on can make it degrade and lose activity.Therefore,in recent years,using the microcapsule technique to process astaxanthin is gradually valued.Using the Haematococcus pluvialis as the raw materials,this subject adopts organic solvents to extract astaxanthin,and embeds the gained astaxanthin oleoresin with microcapsule through the spray drying process.This subject explores the storage stability and external antioxidant capacity of the gained products,and provides theory reference for the functional development of the microcapsule products.The conclusions of the experiment are as follows:1.Extract astaxanthin through the organic solvent method: through inspecting the influential factors including the ratio of material and water,extracting times in the extracting process,get the best extracting condition is that: extract it under the ratio of 1:15 of Haematococcus pluvialis powder and ethyl acetate twice,and the extracting time of every time is 60 minutes.The gained oleoresin is steamed out the residual solvents through the water steaming drying method.By using the above extracting condition,we can get the conclusion that the astaxanthin content and average water content of the final product is respectively 9.81% and 49.55%.2.The optimizing of the microcapsule technique conditions of astaxanthin: select gelatin and maltodextrin as the shell materials,and through the single factor and response surface experiment,optimize the conditions of the emulsion formulation and spray drying process,then finally confirm the best process condition is: ratio of shell materials(maltodextrin: gelatin)is 2.1:1;the ratio of shell core is 5.9:1;the ratio of emulgators(glycerin monostearate: sucrose fatty acid ester)is 1.1:1;the solid content is 14%;emulgators volume of addition is 3%;emulsifying shearing speed is 10000r/min;the shearing time is 10 minutes;embedding rate of astaxanthin microcapsule is 38.02% under the condition of spraying and winding temperature being 180?.3.The product of astaxanthin microcapsule is orange,and the particle surface is smooth with uniform size.Part of them are sunk and not closed.The product quality is overall good.The water content of the microcapsule product is 3.34%;solubility is 96.24%;density is 0.29g/cm3;the angle of repose is 33.05°;the liquidity is good and is not easy to cake or mildew,which benefits for the storage of the product.4.The research of the storage stability of astaxanthin microcapsule: through the contract experiment of astaxanthin microcapsule and astaxanthin oleoresin,find that lighting,oxygen and temperature can be easy to cause the degradation of two kinds forms of astaxanthin,and among them the reservation of astaxanthin in microcapsule is evidently higher than oleoresin.The degradation speed of two forms of astaxanthin in oxygen environment is evidently higher than in high temperature and lighting condition,and the reservation in vacuum environment of low temperature and protection from lighting is highest.5.External antioxidant experiment of astaxanthin microcapsule: astaxanthin microcapsule has scavenging action to hydroxyl radical,DPPH and ABTS,and the scavenging ratio and sample concentration is directly proportional,and the whole oxidizability is higher than VC.Through calculating the value of IC50,it reflects that the scavenging ability of astaxanthin microcapsule to the three kinds of radicals from more to less is: ABTS> hydroxyl radical(-OH)>DPPH,and the result can provide theory basis for the application of astaxanthin microcapsule in all fields.
