In recent years,abuse of amphetamine-type stimulants has been on the rise in the global.The reagent(Ag and Ab)used in company's amphetamine colloidal gold strip depended on imports.The cost of reagent was higher.Therefore,self-development of amphetamine antigen and antibody has important economic value and social benefit.Carboxyl group was added on amphetamine N-site and phenyl,and combined with carrier protein to prepare the antigen.Then the antigen was used as immunogen to prepare specific monoclonal antibody.At the same time,the production process was optimized and confirmed.Finally the performance of antigen and antibody was confirmed by the amphetamine colloidal gold strip.The results showed that amphetamine antigen can be prepared by the process as follow: first amphetamine was reacted with succinic anhydride on the phenyl,then added DCC and NHS,combined with BSA(5mg/ml)in pH 7.4 buffer under low temperature.And then the amphetamine antigen was used as the immunogen to prepare the required amphetamine monoclonal antibody.The antibody production process as follow: the density of cell in culture was 10×104,the density of cell in ascites production was 80×104,the strain of mouse was F1,the purification method was Octanoic acid-ammonium sulfate precipitation.The performance of antigen and antibody met the requirement of amphetamine colloidal gold strip.The reagent that self-developed not only reduced the dependence on imported reagent,but also greatly reduced the cost because its cost was less than 30% of the imported reagent.
