Recently,gold nanoparticles(AuNPs)have been widely used in a variety of biochemistry analysis and biomedical detection techniques,and it produces a series of high selectivity and high sensitivity biochemistry analysis detection methods.In this work,we used single-stranded DNA as a stabilizer to modify the gold nanoparticles by self-assembly technique(Au-S convalent bond).Then ruthenium(?)complexes and coumarin 343 modified by complementary sequences of ssDNA was functionalized on gold nanoparticles.These functionalized gold nanoparticles could be used to detect deoxyribonuclease ?(DNase ?).The sensing of glutathione was also studied on the base of the quenching properties of gold nanoparticles.In chapter one,the nature of the gold nanoparticles,the principle of luminescent quenching,the synthesis methods of gold nanoparticles,and the applications in the chemical and biological sensing were introduced.Then the luminescence mechanism of ruthenium(?)complexes and the applications in the biosensing field were introduced.In addition,the application and detection methods of deoxyribonuclease ?were introduced.The effect of glutathione on human health and the main detection method were introduced.In chapter two,a kind of ruthenium(?)complexes-functionalized gold nanoparticles and a kind of coumarin-functionalized gold nanoparticles by the connection of double stranded DNA(dsDNA)was designed and synthesized.The luminescent materials were quenched due to the luminescent quenching properties of gold nanoparticles.The luminescence would be restored after the double stranded DNA(dsDNA)was cut off by deoxyribonuclease ?(DNase ?).Based on this mechanism,the deoxyribonuclease ?(DNase ?)activity in vitro could be detected by the fluorescence assay,and its biological sensing ability was studied.The effect of the different length of dsDNA on the luminescence quenching efficiency and sensing ability was also studied.In chapter three,a kind of ruthenium(?)complexes with carboxyl groups was synthesized.Based on the luminescence quenching properties of gold nanoparticles,the colorimetric method and fluorescence essay were used to detect glutathione(GSH).Ruthenium(II)complexes with carboxyl groups and coumarin 343 were compared in the sensing of the glutathione(GSH).
