The Application Of Hydroamination-Lactamization Cascade Reaction In The Synthesis Of Natural Products And Their Analogues

This dissertation towards the application of hydroamination-lactamization cascade reaction in the synthesis of natural products and their analogues,which consists of the following two parts.In the first part,we accomplished the synthesis of camptothecin analogues.Based on our previous work,we searched and optimized the condition of hydroamination-lactamization cascade reaction and applied it in the synthesis of camptothecin analogues through reasonable design.We introduced alkyne to the lactone fragment,and finally got the precursor of camptothecin analogues through Sonogashira reaction.In the end we applied the cascade reaction in the synthesis of one new type of camptothecin analogues.In the second part,we completed the total synthesis of Nauclea alkaloids.We applied the hydroamination-lactamization cascade reaction in the total synthesis of indolopyridine alkaloids,and further investigated the universality of the cascade reaction in the meantime.Finally,we finished the total synthesis of angustidine,neonaucline and 20-bromonauclefine in good yield.Further research on the activity of these natural products and their analogues we synthesized is under way.
