In recent years,as China's socio-economic development and people's living standards improve, people pay more and more attention to upscale health food.Fattening beef is a favored one of them.Consumer demand for fattening beef are constantly growing.Fattening beef has so many fine features just like high tenderness, high juiciness and almost have no chewing residue. Therefore, fattening beef have a very high economic value. In the actual production process, fattening process requires large amount of energy and resources, but the quality of beef from the cattle after the same fattening process varies.Top products accounted for only a tiny proportion. This has resulted in a waste of manpower and resources, reducing the competitiveness of enterprises. So we need to find appropriate solutions to address these issues.Marbling is a main determinants of beef quality, and it is an important economic trait in beef industry, intramuscular fat(IMF)has always been considered as the main factor to affect marbling.Intramuscular fat is a complex trail,it is associated with many genes. Most of the studies have been published were focused on one or few genes, and that would miss some important biological information. To find the missing information,we dig genes which related to IMF in whole-genome wide by whole-genome resequencing. In this study,58 cattle were involved in this study,12 of them were sampled as two groups(R01.high levels of IMF %;R02.low levels of IMF %)to resequencing the whole-genome.A total of 56 GB high quality sequence data was generated by High-throughput sequencing on a HiSeq 2500 platform.Map the sequence data to the Btau-4.6.1 database,80.48% obtained reads in R01 and 81.17% in R02 were successfully mapped to the reference sequence, and 98.45% of the reference sequence was covered by the reads on average, the figure in R02 is 97.71%.And both of R01 and R02 sequence depth was 7X.SNP,InDel and SV were also identified in this study, after that we did the association analysis between R01 and R02 has been down, finally 25 genes was identified as the genes we wanted and was annotated by COG,GO,Swisport,NR and KEGG.Among these annotation,GO annotation has comprehensive information about these 25 genes. In published research, most of these genes were reported related to other characters about cattle,in this study we found them are related to IMF.So we catch the missing information, for further research, our team should have verify them one by one.
