Synthesis Of Dihydrothiophene Compounds Bade On [3+2] Cycloaddition Of D-A Cyclopropane And Thiourea

This paper mainly studies two aspects:1.Synthesis of Dihydrothiophene CompoundsDihydrothiophene compounds are important antibacterial agents and therefore have important uses in pharmaceutical activity.For example,thienopyridone compounds are small molecule inhibitors that inhibit phosphatase activity and prevent infinite reproduction of cells.Treatment of tumor a potential drug molecules,so the study of thiophene compounds synthesis is of great significance.In this paper,dichloroethane as solvent,alkali as additive,Lewis acid under the reaction temperature of 90 oC,cyclopropane and thiourea [3 +2] ring addition reaction to generate dihydrothiophene,the reaction has The higher yield(up to 90% yield),the substrate has good universality(21 examples),the product structure by 1H-NMR,13C-NMR,HRMS characterization.The method is simple to operate,has a very high atomic utilization,solvent and catalyst cheap.2.1,2,3,4-tetrazine compounds1,2,3,4-tetrazine compounds have important applications not only in terms of materials but also in biological activity,particularly as prodrugs with antitumor drugs.In this paper,methylene chloride as solvent,at room temperature,the base catalyzed,?-halogenated acylhydrazone and azodicarboxylic acid diester ring addition reaction to generate 1,2,3,4-tetrazine compounds,the reaction(21% yield),the products were characterized by 1H-NMR,13C-NMR and HRMS.The reaction conditions are mild and the catalyst is cheap and readily available,and the reaction can be carried out on a large scale,which provides a reliable method for the synthesis of tetrazine compounds.
