Trace elements in human serum are closely related with health of people.Consequently,the accurate determination for trace elements in serum is very significant.In recent years,inductively coupled plasma mass spectrometry(ICP-MS)equipped with dynamic reaction cell has prevailed as one of the most suitable methodologies for detection of biological specimens such as blood,serum and urine.Thus,how to establish the method with DRC-ICP-MS to quickly,accurately and simultaneously determinate multi-elements is really necessary.In order to solve the spectral interferences within the process of measurement,the study on working principle and ion-molecule reaction of collision/reaction cell is rather necessary.Meanwhile,building the determination method for human serum trace elements is based on above study results.The detection limits of method are in the range of 0.001 ?g/L to 4.621 ?g/L,the quantification limits are in the range of 0.003 ?g/L to 13.863 ?g/L,the spike recovery for elements fall within 100±10%.Meanwhile,good agreement with the recommended values is obtained for all the elements in this study.And this determination method is simple,fast and reduced the necessary patient sample volume to 2mL.At the same time,it could be employed to the monitor of multi-elementary concentration in human blood serum.Evaluation of measurement method using methodologies means including precision,linearity,determination limits,quantification limits and spike recovery aims at analyzing the accuracy and precision for the method.Besides that,the analyses are further evaluated with ClinChek? serum reference material and comparison with the results and reference values directly demonstrates the reliability of the built method.While instrument is optimized,pre-treatment method might significantly influence the determination results.Due to a much ?lighter? matrix in the serum than whole blood and ?heavier? matrix in the serum than water or urine,the choice of pre-treatment is very significant for the results of determination.Consequently,the same serum sample is treated by various dilute solutions and the linearity,determination limits,quantification limits,spike recovery and the analyzing results of reference material are obtained through DRC-ICP-MS.With the considering above factors which are reached using various diluents,the most suitable diluents for each isotope are concluded.The results show that dilute solution only including HNO3 is best dilution for analyzing As,Cr,Mn and Co.And dilute solution containing HNO3 and Triton should be used for determination of elements Cu,Zn,Cd and Tl.Besides,dilute solution including Tetra-n-butylammonium hydroxide,Triton X-100 and EDTA is suitable for analyzing elements Al,V,Mn,Fe,Cr,Co,Sn and Pb in human serum.
