With the rapid development of China's economy,the living standards of our people have been greatly improved.Compared with the past,the Chinese people's diet and diet structure have undergone tremendous changes,meat products become regular members of our dinner table,and with the accelerated pace of life,fast food has become the ma:in way people eat.This series of changes have led to a significant increase in people's diet intake of fat,the incidence rate of hyperlipoidemia,cardiovascular and cerebrovascular disease are also greatly increased.Dyslipidemia is an important factor leading to this series of diseases,it refers to the body of triglycerides,total cholesterol,high density lipoprotein and low density lipoprotein and a series of abnormal indicators.Gynostemma pentaphyllum contains many pharmacological activities:anti-hypertension,lipid-lowering,and myocardial protection.In this study,we investigated the effect of Gynostemma pentaphyllum on fat accumulation in HepG2 cells.In order to study the effect of Gynostemma pentaphyllum on the accumulation of fat accumulation in HepG2 hepatocytes,the effect of Gynostemma pentaphyllum on hepatocyte fat accumulation was evaluated by setting up the model of HepG2 hepatocyte fat accumulation in vitro.The main research contents and conclusions are as follows:The effects of five gypenosides on the viability of HepG2 cells were screened by MTT one-step detection.Compounds SL-1 and SL-2 were identified as suitable samples and the appropriate concentration of administration was in the range of 10 ?g/ml to 100 ?g/ml.The gypenosides SL-1 and SL-2 were prepared from Gypenosides by repeated column chromatography.Establishing the fat accumulation model of HepG2 cells by oleic acid induction.Intracellular lipid droplets,total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-c)and other aspects were used to evaluate the lipid-lowering effect of gypenosides.The results showed that Gypenoside SL-1 and SL-2 could significantly decrease the intracellular lipid accumulation and significantly decrease the content of TG and TC in the cells.100 ?g/ml Gypenosides SL-1's scavenging rates of TG and TC were 54.8%and 52.7%.Gypenoside SL-2's scavenging rates of TC and TG were 78.3%and 80.2%.Moreover,high concentrations of SL-1 and SL-2 also have very good effects on HDL and LDL.Thus,the two kinds of gypenosides have good in vitro lipid-lowering activity,a better prevention of fatty liver effect.
