| Based on the self-assembly ability of Fmoc-FF,supramolecular hydrogels were obtained through the self-assembly of Fmoc-FFH,Fmoc-FFR,Fmoc-FFD,Fmoc-FFE,Fmoc-FFS or Fmoc-FFC in the combination of metal ions or gold nanoparticles to mimic the activity of natural hydrolase and oxidordeuctase.The structures and morphologies of artificial enzymes were characterized by TEM,AFM,size analysis,CD?FT-IR and so on.The conclu sions can be drawn as follows:1.Metal ion-combined peptide self-assembly to construct artificial hydrolase: Different metal ions were introduced to the self-assembly of Fmoc-FFH,a series of hydrolase model(SA-H?SA-H-Ca2+?SA-H-Ba2+?SA-H-Co2+?SA-H-Ni2+?SA-H-Zn2+ and SA-H-Cu2+)were obtained.Studies showed that these artificial hydrolases were composed of nanofibers with the mixed secondary structure of ?-helix and ?-sheets.It was proved that the activated or inhibited activity of artificial enzyme was affected by the assembly degree which depends on the type and concentration of combined metal ion.2.Gold nanoparticle-combined peptide self-assembly to construct artificial oxidordeuctase: Gold nanoparticles(Au NP)were introduced to the self-assembly of different tripeptides(Fmoc-FFH?Fmoc-FFR?Fmoc-FFD?Fmoc-FFE?Fmoc-FFS? Fmoc-FFC)to form a series of reductase models(SA-H-AuNP?SA-R-AuNP?SA-D-AuNP ? SA-E-AuNP ? SA-C-AuNP ? SA-S-AuNP).AuNP promote the self-assembly of peptides that containing alkaline amino acid,but restrain the self-assembly of peptides that containing acidic amino acids(H,R),the polar amino acid which is uncharged cannot be influence by the gold nanoparticles(D,E).On the other hand,the nanofibers consist of peptide with different amino acid and AuNP show different catalytic activities,SA-D-AuNP and SA-E-AuNP have a higher activity than SA-H-AuNP and SA-R-AuNP,SA-C-AuNP and SA-S-AuNP lost their activities to some degree.Therefore,the activity of the artificial oxidordeuctase can be adjusted through changing the peptide type. |
