| Porous starch is a kind of medicinal carrier material which has attracted much attention in recent years because of its excellent adsorption performance.It often used to adsorb the purpose of drugs.Dextrates are used as a diluent to add to the drug.It can also be used as a sweetener.Based on the preparation of porous starch by hydrolyzing the starch,this paper further prepared pharmaceutical excipients Dextrates by using the remaining saccharification liquid.The effects of starch enzymatic hydrolysis on the structure and adsorption properties of porous starch were investigated.And the preparation process of Dextrates was studied.The main points are as follows:1.We selected the adsorption performance as the targets for optical the processes.And we investigated the effects of enzymatic time,enzymatic temperature,enzymatic p H and enzyme content.After analysis,we found the optimal condition is the enzyme dosage 0.5%(w/w),p H is 5.0,temperature is 50℃,treat 12 h.The final oil absorption rate of porous starch is up to 66.9%.2.In order to study the rheological properties of the glucose solution,the effects of concentration and temperature on the viscosity of the solution were investigated.And the empirical formula is used to fit the model.The results found in a certain range the effects of concentration and temperature on the viscosity are consistent with the index model.The change of viscosity is considered to be influenced by the formation of hydrogen bonds to a certain extent by using 1HNMR technique.3.This paper investigated the crystallization behavior of high concentration glucose solution by SEM and XRD.By observing the crystallization of the solution at different concentrations we found that the solutions generated no products when the concentration is smaller than 65%(w/w).And when the concentration is greater than 60%(w/w),the solution is almost completely crystallized.The spherical particle are connected together to form a dense structure at 75% and 80% concentration.In the process of crystallization of glucose,the role of viscosity plays a leading role.The movement of sugar molecules in the solution are blocked at high viscosity,resulting the more irregular crystal.So in the crystallization process using 65% concentration as the crystallization concent ration.By measuring the rheological properties of the solution before and after crystallization,it was found that the solution was transformed from the initial Newtonian fluid into a non-Newtonian fluid with shear thinning.4.With evaluating the effect of different granulation methods on the structure of Dextrates,we found that fluidized bed spray drying is most suitable for granulation of Dextrates.At the end of the experiment,the process stability of Dextrates was tested.The results proved reliable,stable process,and can be used for industrial production. |
