In this research,Se-enriched Pleurotuseryngii were systematic studied which included:isolates screening,fermentation conditions optimization,acute toxicity test and the pharmacologic experiments on the effects of improving immunity and antioxidation of Se-enriched P.eryngii powder.The method was proper and the main contents and conclusions are listed as following:16 strains of P.eryngiiwere screened using solid and liquid medium contains selenium.Results:the growth of Pr06,Pr10,Pr12 and Pr15 were better than other isolates in solid medium and the colony diameter were larger;the growth of Pr06,Pr10 and Pr15 grew better than other isolates in liquid medium,fruiting bodies weight were bigger.The 16 Pleurotus strains' growing cycles are 47.86-67.41 days.Fruiting body production is 32.15g-81.2574g/bottle,the difference is significant.The 16 Strains of were taken into account the comprehensive assessment of the growth time position,fruit bodies weight 2 factors,and the result is that Pr10 were selected as the experimental isolate.The effects of different fermentation conditions for mushrooms deep fermentation.The results showed that the culture temperature 25?,initial pH 6.0 naturally,inoculum 12%,250mL liquid volume 100mL shake flask conditions,mushrooms submerged fermentation of biomass and Se-content of mycelia production max.Also investigated different carbon and nitrogen sources for the fermentation of mushrooms,combined with Se-content and the actual industrial production,agricultural waste select glucose and soybean meal is a suitable carbon and nitrogen sources,actual production has a certain significance.Further orthogonal experiment to determine the best mushroom submerged fermentation medium consists of:2%glucose,soybean meal 3%,KH2PO4 0.025%,MgSO4.7H2O 0.05%,based on the best medium to validate the resulting combination of the above analysis,the results fungus balls dry weight 10.15 mg/mL,compared to the previous optimization improves 29.13%.The maximum Se-content was 2138.52 ?g/g,compared to the previous optimization improved 30.21%.Contents of major chemical components and inorganic elements in Se-enriched P.eryngii powder were determined.The contents of polysaccharides,fat,protein,and moisture reached 22.57%?21.42%?3.76%?4.24%,respectively,and the contents of heavy metal were lower than the standard.The content of Ca was high.The mice acute toxicity of Se-enriched P.eryngii was tested.The result showed that there were not die and pathological changes about drug groups and control group after maximally tolerating in two weeks.The MTD of Se-enriched P.eryngii powder by ig administration was greater than 6267 mg/kg to mice,and the se-enriched P.eryngii powder was belongs to non-toxic substance according to the grade standards of mice acute toxicity byiq.The effects of improving immunity,and antioxidation and anti-tumor of se-enriched P.eryngii powder were tested.The results showed that organ index of Se-enriched Pleurotus eryngii powder groups were significantly higher than that in distilled water group,Na2SeO3 group and normal P.eryngii group;The extracts with pure water,75%ethanol and 80%methanol from the Se-enriched P.eryngii powder could decrease 72.69%,68.31%and 69.72%of 0.2 mg·mL-1 DPPH free radicals,significantly higher than those from normal Pl.eryngii power and mycelia grown in Se submerged culture;Plasma and liver GSH-Px,SOD activity of Se-enriched P.eryngii powder groups were significantly higher than those of distilled water group,Na2SeO3 group and normal P.eryngii group,and MDA content in plasma and liver were lower than those of the other groups.
