Polysaccharides are the main active substance of G.Frondosa,which has significant pharmacologic actions of anti-tumor,anti-HIV viruses,immunoloregulation,anti-radiation and hepatitis treatment.This paper mainly studies the extraction,separation and purification of polysaccharides from G.Frondosa and is divided into three parts.Firstly,conditions of microwave extraction method and pressurized solvent extraction are optimized by the orthogonal method.The optimum extraction conditions are the extraction duration of 15min,solid to liquid ratio of 1:40(g:mL),the extraction temperature of 90? and the extraction times of twice.Under these conditions,the actual extraction rate is 3.731%.The optimum extraction conditions of pressurized solvent extraction are the extraction duration of 15 min,the extraction pressure of 90bar,the extraction temperature of 90?,for three times.Under these conditions,the actual extraction rate was 3.018%.Secondly,conditions of sevag method are optimized by the orthogonal method.The optimum extraction conditions are that chloroform:n-butanol(v:v)was 5:1,sample:reagent(v:v)was 5:1,for three times;activated carbon discoloration,hydrogen peroxide decoloration and five macroporous resins are investegated through comparing protein removal rate,decoloring rate and polysaccharide preserving rate;Finally,polysaccharides from G.Frondcosa by ultrafiltration are divided into three parts:the>0.2?m part,the 50K-0.2?m part and ?50K part;and the polysaccharides are obtained by DEAE-Sepharose Fast Flow ex-change resin and Sephacryl S-Series gelatin.
