Flavonoids from bamboo leaves has multiple biological effects,such as anti-oxidation,anti-free radical,anti-inflammatory,anti-bacteria,anti-virus,antitumor,and prevention of cardiovascular diseases.The aim of the dissertation is to establish a practical method for the isolation and purification of flavone compounds from bamboo leaves to get the high purity and security flavonoids,and improve its lipid solubility by entrapment method,and evaluate the antioxidant activity in the oil.First,the separation and enrichment of flavonoids from bamboo leaves were studied on polyvinyl polypyrrolidone(PVPP).In order to optimize parameters of separation,dynamic adsorption and desorption experiments were carried out on the columns packed with PVPP.The optimal conditions were:10 mg·mL-1 crude extract solutions at the flow rate 0.33 mL·min-1,and the optimum diameter height ratio was 1:1.9;the optimal solution for the desorption was water with 1BV,70%ethanol with 800 mL at the flow rate of 0.33 mL·min-1.Under this condition,the purity of flavonoids was above 90%with segmentation receiving 80?560 mL eluent and was above 95%with 80-480 mL eluent,and the recovery rate was 63.5%.The regenerative PVPP was used to purify flavonoids in the optimal conditions,and the purity of flavonoids was above 90%with segmentation receiving 80?560 mL eluent and was above 95%with 160?480 mL eluent,and the recovery rate was 61%.Second,fat-soluble flavonoid-embedded objects was prepared with glycerolmonolaurate(GML)and Span 80.Single Factor Experiment was carried out with the edible vegetable oil as blank,the absorbance of the oil with flavonoid-embedded objects in the 760 nm as the indicator.The smaller of the absorbance,the smaller of the turbidity.The optimal conditions were as follows:emulsification time 30 min,emulsification temperature 50 ?,95%ethanol 10 mL,Span 80 0.4%,emulsification speed 1700 r?min-1,rotary evaporation temperature 50 ?,rotary evaporation speed 120 r·min-1,the drug loading 11.1%.Under this condition,the absorbance of the oil in the 760 nm is 0.027A when the flavonoids concentration is 0.02%.Third,fat-soluble flavonoid-embedded objects were prepared with monosterin and Span 80 in the optimal conditions.The effect of flavonoids purity and the solids content for the flavonoid extract on fat-soluble was investigated.The results showed that the turbidity dicrease with the increase of the flavonoids purity and the decrease of the solids content.The effect of the organic solvent of flavonoid extract on the fat-soluble was investigated.It revealed that removing organic solvent is good to improve the solubility of flavonoids embedded ojects in the oil.Last,the antioxidant activity of flavonoids embedded ojects in the oil was determined by titration method and compared of the peroxide value of centrifugation and not centrifugation oil solution.The results showed that the peroxide value of the not centrifugation oil solution with flavonoids embedded oject was lower than centrifugation oil solution,and the peroxide value was the least when the flavonoids concentration was 0.02%.
