The bacterial cellulose(BC)'s three-dimensional structure properties such as porosity will significantly affect the cell behavior when it as the cell matrix material. In addition, the BC membrane water and water release rate was closely related to wound healing. In the biosynthesis process of BC, oxygen is the important influence factors in the aerobic fermentation process. It is not only closely related to cell growth and metabolites, but also has important influence on the bacterial cellulose membrane microstructure. In this study, bacterial cellulose membrane were produce by Gluconacetobacter xylinus with different oxygen-enriched air. By detecting the difference of macroscopical structure of BC and the key intracellular metabolites components of G. xylinus, determine its effect on the growth of G. xylinus and bacterial cellulose synthesis. The results will help us to understand the mechanism of how oxygen regulating BC biosynthesis and assembly and to build a new production method for personalized BC products. In the research of the liquid medium which were filled with oxygen-enriched air, the grow of bacteria were influenced by increasing the content of the oxygen in the medium. The exponential phase will .be obvious influence on the growth of bacteria. When the medium filled with oxygen-enriched air(the oxygen content: 20%, 40%, 60% and 80%), the cell volume were increased by 610%, 470%, 260% and 300%.In the research, the concentration of oxygen in the oxygen-enriched air had great effect on the physical chemical properties of BC. The differences of the mechanical properties and porosity were obvious. With the increase of oxygen content in the oxygen-enriched air, the texture and porosity of BC has changed significantly. When the oxygen concentration in the oxygen-enriched air reached 60%, porosity of BC is the largest. The metabolism of G. xylinus had an obvious effect when the medium filled with oxygen-enriched air. The cell grow up were faster when the medium filled with oxygen-enriched air. It make the consumption of glucose increased. The residual glucose always less than the which in the medium didn't filled with gas. When the medium filled with oxygen-enriched air(the oxygen content: 20%, 40%, 60% and 80%), the cell volume were decreased by 27.8%, 61.1%, 4.2% and15.3%. The accumulation rate of glucose acid were faster at the beginning of the cultivation when the medium filled with oxygen-enriched air. It also accelerate the consumption of glucose acid. It also make the cell volume increased under the condition of same glucose consumption.
