Fluorescence analysis method is developing very rapidly,drawing more and more attentions for its simple operation,fast response time,low cost,inexpensive equipment in the last ten years.Because of good optical properties,high molar absorption coefficient and quantum yield,porphyrin compounds with conjugate ring system are often used as a kind of sensor component,which has become a research hotspot at present fluorescence analysis method.Copper is an indispensable metal nutrient for metabolism in the human body as well as in the environmental cycle,which is vital to the health of all living things.The imbalance of copper can lead to tissue injury and disrupt the function of normal cells,causing a series of diseases in living things.Acid phosphatase?ACP?,an abundant digestive enzyme,is widely found in mammalian body fluids and tissues.The imbalance of ACP level can also cause many diseases.Some kinds of ACP can be diagnostically useful as serological and histological diseasemarkers and be useful in the pathophysiology of theassociated diseases.Cyanide,as a kind of highly toxic substances,has been widely applied to all areas of our lives,long-term exposure in the environment of cyanide pose a growing threat to our health.In this dissertation,we synthesized two fluorescent probes for the detection of copper ion,cyanide and acid phosphatase.The main conclusions are as follows:1?Preparation and properties of fluorescent probes Por-1 and Por-2 for the detection of Cu2+We designed and synthesized two fluorescent probes Por-1 and Por-2,which were characterized by NMR,UV,and fluorescence analysis method were used for quantitative detection of Cu2+.The results showed that both of the two probes can detect Cu2+with high selectivity and sensitivity.Based on the analysis of the experimental data,the mechanisms of the probes for Cu2+were proposed.The detection limit of probe Por-1 for Cu2+was about 1.04?M and probe Por-2 for Cu2+was about 0.123?M.The interference of other metal ions on the detection of Cu2+was negligible.The probe Por-2 contains multiple phenolic hydroxyl groups,which greatly increases the water solubility and biocompatibility,and provides the possibility for the detection of Cu2+concentration in water samples and blood.2?Study of biosensor Por-2 for the detection of acid phosphatase?ACP?We used water-soluble fluorescent probe Por-2 containing phenolic hydroxyl groups for quantitative detection of ACP activity by using UV and fluorescence analysis method.The results showed that the probe can detect ACP activity with high sensitivity,and the detection limit of ACP was about 1.73×10-8 M.Through the data analysis of the concentration titration experiment of adenosine triphosphate ATP and ACP,we proposed the detection mechanism of ACP,which was a process of fluorescence"off-on-off".The sensitivity analysis of ACP activity provides a feasible method for detection of ACP in biological system.3?Preparation and properties of fluorescent probePor-2 for the detection of CN-We used synthetic water-soluble fluorescent probe Por-2 containing multiple phenolic hydroxyl groups for qualitative and quantitative detection of CN-by using UV and fluorescence analysis method.The results showed that the probe Por-2 can detect CN-with high selectivity and sensitivity,the detection limit of CN-was about 0.42?M.The interference of other anions to CN-is very insignificant except H2PO4-.The water soluble fluorescent probe Por-2 provides the possibility for the detection of CN-concentration in water samples.
