Sporidiobolus pararoseus JD-2 belongs to genus Sporidiobolus(family Sporobolomycetaceae)and is found rich in lipid and carotenoid,which could be developed into functional food.The nutritional components and hypolipidemic effects of S.pararoseus oil were analyzed in this paper to lay the groundwork of further exploitation and utilization of S.pararoseus oil in the future.The method of cell-wall breaking and lipid extraction of S.pararoseus JD-2 oil was firstly optimized in this study.An optimized breaking method was developed with the minimum destroy on nutritional components,following with the analysis of its component.Ten mice were randomly selected from 60 mice as control group(BC)and the rest 50 mice were fed with a high-fat diet to be a dyslipidemia model.The effect and mechanism of blood lipid metabolism in mice with dyslipidemia were studied by the method of intragastric administration in mice.After 4 weeks,The 50 mice were randomly divided into 5 groups: high-fat model group(HM),low dose of S.pararoseus oil(SL),middle dose of S.pararoseus oil(SM),high dose of S.pararoseus(SH)and doxicon group(PC).The mice were sacrificed after eight weeks and the concentration of total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C)and low density lipoprotein cholesterol(LDL-C)in serum were measured.Liver and fat histological analysis was performed by microscopy.The mRNA expression of SREBP-1c?FAS?ACC and CPT-1 in liver were measured by RT-PCR.The results indicated that:(1)High pressure homogenization,its cell-wall breaking rate as high as 51.19%,was a harmless method.It destroyed nutritional components barely.(2)S.pararoseus oil had a lot of unsaturated fatty acid and the concentration of oleic acid was 71.46%.There were carotenoid(0.41 g/kg),squalene(1.46 g/kg)and ergosterol(3.13 g/kg)in the oil.(3)Compared with the HM group,the body weight,BMI,epididymis fat index of S.pararoseus oil group were significantly decreased.And heart,liver,kidney index were increased.The oil was harmless for organs and maybe can repair and protect them.(4)S.pararoseus oil can increase SOD,T-AOC,GSH/GSSG level and decrease MDA level.The oil can increase total oxidation resistance,reduce the degree of lipid peroxidation,relieve the oxidative stress damage.(5)S.pararoseus oil can decrease serum TC,TG,LDL-C level.The oil reduced effect of TC and LDL-C is greater than doxicon and its reduced effect of TG is less than doxicon.The oil can restore morphology of liver cell,improve liver fat accumulation phenomenon,but also reduce the fat cell volume,thus controlling the growth of weight and body.(6)RT-PCR result showed that S.pararoseus oil can decrease expression of cholesterol synthesis related genes SREBP-2,HMGCR and increase expression of catabolic related genes LXR?,CYP7a1 thus decreasing TC.The oil can decrease expression of fatty acid metabolism related genes ACC,SREBP-1c,increase expression of FAS,CPT-1 thus decreasing TC.(7)Correlation analysis result showed that liver antioxidant enzymes has a certain degree of regulation of cholesterol metabolism related genes.Better antioxidant effect,lower serum lipid levels.Above all,S.pararoseus oil had beneficial effects on the control of serum lipid and it had more beneficial effects on the control of TC,LDL-C than doxicon.LDL-C was the main goal of hypolipidemic therapy.Obviously,S.pararoseus oil was better than doxicon in this respect.
