3-Amino-1,2-propanediol is an important pharmaceutical intermediate.The domestic product usually has some defects such as poor appearance,lower purity and higher impurity which is caused by the by-product 2-amino-1,3-propanediol.Hence,the domestic 3-amino-1,2-propanediol is difficult to meet the requirements of manufacturers which has to turn to the abroad product.In our research,3-chloro-1,2-propanediol was synthesized from 2-(chloromethyl)oxirane by acidic ring opening hydration reaction.The method of feed and reaction was optimized.By using molecular distillation technology for the final purification,the yield has been greatly improved compared with reported results.The subsequential ammoniation of 3-chloro-1,2-propanediol with ammonia was under certain catalyst.After the screening of the catalyst,the temperature,the time,the ratio of the materiels and the pressure of the reaction,the optimal conditions for the reaction were obtained.After the workup and vacuum distillation,the final 3-amino-1,2-propanediol was achieved as a colorless viscous transparent liquid.The procedure was superior to known methods with lower appearance of byproduct and higher yield of the 3-amino-1,2-propanediol.Meanwhile,based on the published literature,the application of 3-amino-1,2-propanediol in anticancer drug preparation has been developed.The derivatives from the reaction of 3-amino-1,2-propanediol with isocyanates were obtained.Based on the preliminary study on the synthesis of 3-amino-1,2-propanediol,the pilot process of semi-works production of 3-amino-1,2-propanediol was explored.A 200-ton pilot process has been designed,and the material accounting as well as flowprocess diagram has been given.The production conditions of the process is mild,and the time consumption is short,and the efficiency is high.All above would satisfy the production requirements from the manufacturer.The research provided a novel method for the synthesis and manufacturing of 3-amino-1,2-propanediol and their derivatives.
