The enzymatic hydrolysis process of protein bait fromulation using the waste beer yeast was studied in this paper.Four kinds of protease and the optimal application condition were studied to choose the best enzymatic hydrolysis condition.In addition,the bioassay of the protein bait was conducted in the laboratory.The results were as follow:1 The best enzymatic hydrolysis condition of acid proteaseThe single factor experiment was designed by the attractant rate as evaluation index.Through the single factor tests,the optimal application condition were analyzed,such as enzymatic hydrolysis temperature,pH,ratio of protease and hydrolysis time.The results showed that the optimal temperature was 60 ?,the optimal pH was 4.4,the optimal ratio of protease was 0.2%,the optimal hydrolysis time was 22h.The orthogonal experiment was designed by combining the single factor experiment.The results of orthogonal experiment showed that the best enzymatic hydrolysis condition of acid protease was T=60?,pH=4.4,ratio of protease was 0.2%,optimal hydrolysis time was 24h.2 The best enzymatic hydrolysis condition of alkaline proteaseThe single factor tests results showed that the optimal temperature was 50?,the optimal PH was 8.5,the optimal ratio of protease was 0.4%,the optimal hydrolysis time was 26h.The orthogonal experiment was designed by combining the single factor experiment.The results of orthogonal experiment showed that the best enzymatic hydrolysis condition of acid protease was T=60?,pH=8.5,ratio of protease was 0.4%,optimal hydrolysis time was 22h.3 The best enzymatic hydrolysis condition of neutral proteaseThe single factor tests results showed that the optimal temperature was 50??the optimal pH was 7.5,the optimal ratio of protease was 0.2%,the optimal hydrolysis time was 34h.The orthogonal experiment was designed by combining the single factor experiment.The results of orthogonal experiment showed that the best enzymatic hydrolysis condition of acid protease was T=50?,pH=7.5,ratio of protease was 0.1%,optimal hydrolysis time was 34h.3 The best enzymatic hydrolysis condition of papainThe single factor tests results showed that the optimal temperature was 50 ?,the optimal PH was 6.5,the optimal ratio of protease was 0.2%,the optimal hydrolysis time was 26h.The orthogonal experiment was designed by combining the single factor experiment,The results of orthogonal experiment showed that the best enzymatic hydrolysis condition of acid protease was T=50?,pH=6.5,ratio of protease was 0.2%,optimal hydrolysis time was 26h.5 The comparison of enzymatic hydrolysis effect of four kinds of proteaseThe waste beer yeast was hydrolyzed by four kinds of protease under the best enzymatic hydrolysis condition.The result of laboratory bioassay of final product showed that acid protease and papain were better to attract,but combining with other factors such as economic factors and production,the final recommendation was papain.The best enzymatic hydrolysis condition of papain was T=50?,pH=6.5,ratio of protease was 0.2%,optimal hydrolysis time was 26h.
