With the progress of modern biotechnology,the researches of selenium as well as gradually improve the functions have proved that selenium is the key group of proteins required for a variety of human body metabolism,such as antioxidant,reduce inflammation,the production of thyroid hormones and improve fertility.However,in our daily life exposure to various types of natural food,which selenium content is too small,furthermore some difficult to use.As a result,natural foods are unable to meet the daily needs of selenium.In face of the situation that the demand is greater than the quantity supplied,it is very significant to substitute selenium rich food,such as selenium supplement.The safety coefficient of organic selenium is big,not easy to cause poisoning,and high utilization rate,easy to absorb,compared with inorganic selenium,which not only reduces the toxicity but also improve the utilization rate of selenium,in human health to the development direction of selenium,its forward to a major step.In recent years,domestic and foreign literature has shown that selenium can treat or prevent hyperlipidemia by regulating the metabolism of human body.Therefore,after detecting the acute toxicity and genotoxicity of biological selenium prepared in our laboratory,the hyperlipidemic mice were treated with intragastric administration to evaluate its function of reducing blood lipids.The main research contents and results are as follows:1.Kunming mice acute toxicity test with 6~7 weeks of age 300,the biological selenium used modified Karber method on 7 different fermentation conditions were tested,the results showed that the Streptococcus thermophilus and yeast as main strains,YPD as culture medium,the selenium content of fermentation =2000mg/L,No.6,7 of months biological selenium toxicity minimum,No.6 LD50=525.21 mg/kg,No.7 LD50=392.55mg/kg,therefore the two groups carry out follow-up test.2.A total of 40 Kunming mice of 6~7 weeks old were selected and divided into three groups(n = 8 in each group),with each group containing only 5 mice.In the experimental group,the 1/2 and 1/4,1/8LD50 of biological selenium of No.6 and No.7 were selected from the acute toxicity test.The negative control group were lavaged with physiological saline and the positive group were lavaged with intragastricadministration of cyclophosphamide 40 mg/kg by weight.The test of different dose group and cyclophosphamide 40 mg group aberration rate had significant difference(P<0.01);biological selenium is high,medium and low dose group and saline control group showed no significant difference(P>0.05);compared with saline control group,the P value of chromosome aberration rate of cyclophosphamide group less than 0.01,which proved experiment desirable;6 dose biological selenium group and negative group had no obvious statistical difference relatively,so when the biological selenium is less than or equal to 262.61mg/kg of 6 biological selenium and less than or equal to196.28mg/kg of 7 biological selenium,biological selenium was negative.3.Experimental study on treatment of hyperlipidemia mice with biological selenium,100 male animals were selected and divided into eight groups.At first the high fat model was established,then the high,medium,low concentration of selenium was set up in the stomach,and established positive and negative controls.After treatment,found that biological selenium have lipid-lowering effect,and has no significant adverse effects on the liver oxidation index,therefore based on this test,choose a relatively better biological selenium of No.6 for subsequent prevention trials.4.By comparing the effect of three different doses of No.6 biological selenium in the prevention and treatment of the gradient,it was found that the preventive effect was significantly better than the therapeutic effect.The blood lipid,TC: treatment group,total cholesterol lowering ability of medium and low dose group of is weak,there is significant difference with other groups;TG: the difference is not very significant,can be clearly seen in the medium dose group,prevention and treatment group has obvious advantages,but the effect of the prevention group and treatment group is not obvious difference.HDL-C: there was negatively correlated between the index and hyperlipidemia,AS various diseases,which proving in this aspect of Western medicine and biological selenium group were had a good influence.Antioxidant,SOD: there was no statistically significant difference in superoxide dismutase index each group,it can be speculated that there was no significant free radical production.MDA: Western medicine Lipitor produces more harmful MDA,and compared with the prevention group,the value of the treatment group relatively high.GSH-Px: in the treatment group and the prevention group,there was directlyproportionalthe between the enzyme activity and the amount of biological selenium.In general,biological selenium has a certain antioxidant effect on model mice,and in general,the indicators of the middle dose group of the prevention was the best.Taking into account the biological selenium toxicity was small,no obvious genetic toxicity,there was good performance in the case of auxiliary lipid-lowering,as well as no obvious side effects,consequently used medium dose prevention treatment.
