Fluorescence probes and fluorescence technology,which can be used for fluorescence imaging of living cells and model animals,have been developed rapidly in recent years.Azo reductase is an important enzyme in life.The level of azo reductase in tumor is closely related to the degree of hypoxia.The detection of azo reductase levels can be used to evaluate the degree of hypoxia of tumors and further to determine the benign and malignant regions.Therefore,the detection of azo reductase levels in living cells and living tissues by fluorescence probe is of great significance in biomedical and clinical diagnostics.In this dissertation,a fluorescent probe with selective photophysical properties towards azo reductase was designed and synthesized.This probe was successfully applied to azo reductase fluorescence imaging in living cells,Caenorhabditis elegans and lab rats.The main work is as follows:1?I studied the photophysical properties of these fluorophores and found that fluorophore C3 in DMF-PBS buffer(0.02 M,pH 7.4)(V/V = 5:5)The maximum emission wavelength is at 675 nm,in the near infrared fluorescence emission region(650-900 nm).2?I synthesized two new fluorescent probes S1 and S2 for the detection of azo reductase based on fluorophore C3.The fluorescence recognition and fluorescence imaging of azo reductase were studied.The addition of azo reductase led to a emission change from red color to green color in both cases of probes S1 and S2.We use the photophysical properties of more excellent probe S1 as the main research object,The mechanism of probe S1 and azo reductase was studied.The results of probe S1 in biofluorescence imaging(cells,C.elegans and tumor tissue)showed that the probe S1 had good spectral responses towards azo reductase,and it can be used to detect azo reductase in living cells and living tissue.
