Aldo keto reductase AKR1C3 is widely distributed in human tissue,which is closely related with cancers.In this work,we use GreenI SYBR real-time fluorescence quantitative PCR,sandwich ELISA,enzyme inhibitor model to research the relationship between AKR1C3 and liver cells,liver cancer cells,lung cancer cells and colon cancer cells on molecular level,protein level and enzyme properties.The results showed that the concentration of AKR1C3 in liver cancer cells was the highest.AKR1C3 was in low level in the liver cells,colon cancer cells and lung cancer cells.ELISA showed that the target protein in liver cancer cells was 3.2 ng/?g,the liver cells were 0.81 ng/?g,colon cancer was 0.64 ng/?g,the content of 0.68 ng/?g in lung cancer.The model in vitro indicates,indomethacin,sulindac,progesterone,flufenamic acid,methyl jasmonate,2'-hydroxyl brass,cloxazolam could inhibite Aldo keto reductase AKR1C3.Inhibition activities are shown as following: IC50 2'-hydroxyl brass <IC50 progesterone<IC50 sulindac<IC50 cloxazolam <IC50 flufenamic acid<IC50 indometacin <IC50 methyl jasmonate.These results indicate that AKR1C3 is closely related to generation and development of liver cancer.The results of seven inhibitors showed that progesterone can be used to inhibit the activity of AKR1C3 to inhibit liver cancer,and then achieve the purpose of treatment.The research results provide experimental basis for the diagnosis and treatment of liver cancer and related cancers by AKR1C3 level.
