As a new type of simple analytical device,paper-based microfluidic chips,which combines the miniaturization of microfluidic technology and the advantages of paper materials,has become the focus of current attention.It has shown great application potential in environmental monitoring,food quality control and biomedical aspects.This work aims to propose a novel method to fabricate 3D microfluidic paper-based analytical devices,and studies the functional research of paper chips.It can be divided into two sections:Firstly,the advantages,processing,detection methods and applications of paper chips were reviewed.Secondly,the development of 3D paper chips was introduced,and the processing technology and controlling methods of 3D paper chips were highlighted.Finally,the specific application of enzyme-linked immunoassay in paper chip is introduced.The second section is the research report,which consists mainly of three parts.The specific contents are as follows:Part ?,the paper mainly introduces a new method to make 3D microfluidic chips by paper clips.Multiple paper chips were fabricated.The channel verification experiment,reproducibility of the method and performance experiment were explored.Using self-made colorimetric detection device,this approach can be successfully applied to fast quantitative detection of bovine serum albumin and ferrous iron.In a specified range,they both have the better linear relation.The comparisons of the proposed method and other methods were studied,and verified the possibility of our method.Part ?,this paper was described a novel strategy to fabricate the movable valve on the paper-based microfluidic devices to manipulate capillary-driven fluids based on paper-clips method.This method is applied to realize colorimetric immunoassay of carcinoembryonic antigen(CEA).The 3D paper-based microfluidics were fabricated by wax printing method,and integrated together with valves by the rivets.To verify the performance of the movable valve,several different designs of ?PADs were tested andobtained with satisfactory results.In addition,double antibody sandwich enzyme-linked immunosorbent assay(ELISA)experiments on ?PADs were studied,the use of these valves in ?PADs was applied successfully for analysis of carcinoembryonic antigen(CEA),it showed good sensitivity and reproducibility.The CEA in the actual serum was detected by this method,and verified the feasibility of our method.Part ?,the paper was described a novel strategy to fabricate the rotary valve on the paper-based microfluidic devices.A novel method for the detection of alpha-fetoprotein(AFP)in serum was established on a paper chip combining the hydrogen-peroxidase(HRP)-mediated luminol chemiluminescence system.Based on HRP can effectively catalyze the chemiluminescence of luminol-H2O2 chemiluminescence system,the assay of detecting HRP in the sample realized.At the same time,simple chemiluminescence detection equipment was set up.The results showed that there was a good linear relationship between chemiluminescence signal intensity and AFP concentration.The AFP in the actual serum was detected by this method,and the operability of the rotary valve paper chip was proved.
