In this study,the purification effect of halophyte Sesuvium portulacastrum and algae in aquaculture water was carried out,the best density ratio was obtained through optimizing technological parameter.The purpose of the article is to provide basical support of building biological restoration project to restore aquaculture water.Meanwhile,compare the composition and structure of bacteria communities in samples including untreated aquaculture water,comprehensive aquaculture water of the polyculture system of Sesuvium portulacastrum and Ulva lactuca,the surface of Ulva lactuca and the root system of Sesuvium portulacastrum based on Hi Seq sequencing of the V4 region of the 16 S r DNA.Theobject is to explore the influence of the addition of Sesuvium portulacastrum and algaeon the composition and structure of bacteria communitiesin aquacrulture water.The main results were as following:(1)The experiment was made on the removal of nitrogen and phosphorus nutrient by Enteromorpha prolifera,Ulva lactuca and Sesuvium portulacastrum.The main conclusions were as following: The purification effect of Enteromorpha prolifera is not very well.When the stocking density of Ulva lactuca is 2g/L,purification effect isIV the best,and far better than Enteromorpha prolifera in removal effect.When the planting density of Sesuvium portulacastrum is 1.5 ind/L,purification effect is the best.(2)The experiment was made on the removal of nitrogen and phosphorus nutrient by Ulva lactuca and Sesuvium portulacastrum respectively and together.The main conclusions were as following: Ulva lactuca and Sesuvium portulacastrum can improve the quality of aquaculture water,the four experimental groups listed in order of purifying effect: mixed group> Ulva lactuca> Sesuvium portulacastrum>blank control group.The purification effect of mixed group is the best,the experimental result fully demonstrated the purification of the polyculture system of Sesuvium portulacastrum and Ulva lactuca was obvious.(3)The optimal stocking density of the polyculture system of Sesuvium portulacastrum and Ulva lactuca was 6.36 gram and 1 gram per liter,the effect of the polyculture system was obvious.(4)The top 10 phylum of relative abundance of bacteria in four groups are all:Proteobacteria,Cyanobacteria,Actinobacteria,Bacteroidetes,Planctomycetes,Verrucomicrobia,Chlorobi,Firmicutes,TM6 and Chlamydiae.The top 10 genus of relative abundance of bacteria in four groups are all: Synechococcus,Alcanivorax,Mycobacterium,Pseudomonas,Thalassotalea,Marinomonas,Ruggeria and Catenovulum.(5)It shows that bacterial diversity and species richness in four groups of samples are very high and amount of sequencing could cover all the bacteria in the sample groups through the rarefaction curve and Rank Abundance curve.The four groups of samples listed in order of species richness: XWS2>XWS3>XWS1>XWS4.In Alpha diversity index difference between groups analysis,it shows that the species of XWS2 group significantly greater than the species of XWS1group(P<0.05),the species of XWS2 group significantly greater than the species of XWS24 group(P<0.05),there was no significant difference between other groups(P>0.05).Through comparing the shanon index of each group,the four groups of samples listed in order of bacterial diversity: XWS2>XWS1> XWS3>XWS4.The result indicates that the addition ofSesuvium portulacastrum and Ulva lactuca promoted the microbial diversity of aquaculture water.
