Diacylglycerol was a kind of functional oil can reduce the accumulation of visceral fat,weight control,reduce blood fat and other effects.Diacylglycerol was widely applied to all walks of life.diacylglycerol oil had great potential for developmentIn the China market.the method for diacylglycerol by Enzymatic synthesis had advantages: mild reaction conditions,the enzyme had high selectivity and high purity products,low energy consumption.Using soybean oil as raw material to product diacylglycerol,not only can meet the nutritional requirements of raw materials,but also can realize the industrialization.This topic was based on the abundant soybean oil in Northeast china,Research the effect of hydrolysis time,temperature,enzyme dosage,water volume,PH and glycerol molar ratio on diacylglycerol content by lipase In the solvent-free system.The content of glycerol diester was determined by single factor reaction surface method,and the content of glycerol diester was determined by GC-MS(The main component:oleic acid diglycerides and linoleic acid diglycerides).The optimum conditions were as follows:(1)Phospholipase: the amount of water was 25 mL,the hydrolysis temperature was 40?,the amount of enzyme was 700 uL,the reaction time was 12 h.The content of glycerol diester in soybean oil was 16.3%.(2)lipase TLIM: reaction molar ratio(soybean oil: glycerol)5:1,the amount of enzyme 2.0g,temperature 50?,reaction time 16 h.The content of glycerol diester in soybean oil was 17.04%.(3)lipase RMIM: reaction molar ratio(soybean oil:glycerol)5:1,the amount of enzyme 3.0g,temperature 55?,reaction time 16 h.The content of glycerol diester in soybean oil was 18.88%.(4)lipase 435: reaction molar ratio of 5:1,the amount of enzyme 3.0g,temperature 50?,reaction time 12 h.The content of glycerol diester in soybean oil was 17.85%.The effects of distillation temperature,distillation vacuum degree and distillation scraper speed on the content of glyceride diester in soybean oil were investigated by means of molecular distillation.The content of glycerol diester was determined by orthogonal test.The final best process: temperature 130?,vacuum 1.0 mbar,scraper speed 600 r/min.After the three-stage distillation of glycerol diester content of more than 40%.
