The lignans extracted from sesame or sesame meal have been widely used on antioxidants aspects during food processing.However,the extraction rate and biological activity of sesame extracts using current extraction method are not high,which limits its application to a certain extent.A large number of domestic and foreign studies have shown that extracts extracted from sesame seed contained sesamin,sesaminol,sesamol,terpineol and other phenolic substances,and they had good anti-oxidation,antibacterial,anti-inflammatory,lower cholesterol and other biological functions.In this study,sesame meal was selected as raw material.Sesame meal was pretreated with hydrochloric acid and then extracted by alcohol in order to obtain sesame meal extracts with high biological activity.(1)The effects of soaking solution on DPPH free radical scavenging ability of lignans were investigated by single factor experiment.The results showed that the DPPH free radical was scavenged higher by extracts from sesame meal treated with acid soaking,besides the improving effect of sesame meal treated with hydrochloric acid pretreatment is better so that hydrochloric acid was selected to the following study.(2)The effects of acid bubble temperature,liquid ratio,acid leaching time and acid concentration on DPPH free radical scavenging were investigated by single factor and orthogonal experiment so that the optimum acid bubble process of sesame meal extracts was determined.They were liquid ratio 1:15,acid concentration 0.35 mol/L,time 1.5 h and temperature 60 ?.When the concentration of extracts was 1.0 mg/mL,the scavenging rate of DPPH radical was 86.35% and the extraction rate of extracts was 2.65%.(3)The effects of acid leaching on the antioxidative activity of the extracts extracted from the sesame meal under the optimum acid bubble process were investigated by single factor experiment.In addition,the sesamin and sesame meal extract was also added to the peanut oil to determine the effect of the extracts on the antioxidative stability of the oil.The results showed that the antioxidative activity of sesamin and sesame meal extracts on O2-·were poor.Besides,acid leaching can increase the removal rate of DPPH free radical,and the higher the concentration was,the more obvious the antioxidant was.What's more,(4)sesamin and the extracts can increase the oxidation stability of peanut oil,even through it was not obvious.The effect of acid leaching on the oxidation stability of peanut oil was not obvious.(5)The extracts extracted from the optimum acid bubble process were used to study the antibacterial activity.The study added sesamin and sesame meal extracts to Escherichia coli,Bacillus subtilis and Staphylococcus aureus to study the antibacterial effect of the samples on these three common kinds of bacteria.Besides sesamin and sesame meal extracts were also added to the milk rancidity mixed bacteria to explore its practical application in food.The results showed that sesamin and sesame meal extracts had antibacterial activity to these four tested kinds of bacteria,and acid leaching could enhance the antibacterial activity of sesame meal extracts.Besides,the antibacterial activity was more obvious on Escherichia coli.
