Ethyl beta Ethyl ?-apo-8'-carotenoate is an important carotenoid,widely used in food additive [1-5].C5 phosphate is one of the important intermediates of the ethyl ?-apo-8'-carotenoate.Therefore,it is very important to study the routes of C5 phosphate.This paper is based on the laboratory research to find a more suitable route which takes 3-chloro-butene as raw material and through a series of steps such as reaction,elimination reaction and so on.In this paper,the synthesis route of C5 phosphonate,include analyse and evaluate,is briefly introduced.According the analyse,toxic substances as raw material is avoided.Using 3-chloro-1-butene as raw materials,through Grignard reaction and carbonylation synthesis of 2-methyl-3-ethyl crotonate which then takes the next three steps reaction to the synthesis of the product C5 phosphate.The synthetic line has the advantages of short route,convenient reaction,safety and energy saving.Two routes are optimized through single-factor experiment and theory analysis.Including H2SO4 solvent,CO2 in the process of the format of the reaction,hydrolysis acid water solution in Grignard reaction and temperature,catalyst and solvent of the carbonyl reaction.Meanwhile,all compounds use NMR(1HNMR),mass spectrometry(MS)and infrared spectroscopy(IR)spectroscopic methods for characterization.Finally,we sum up the paper,and point out the advantage between Grignard reaction and carbonylation which use 3-Chloro-1-butene as raw material to synthesize C5 phosphonic acid ester...
