Synthetic Studies On Estrone And Its Antigen Derivatives

The synthesis of estrone(1)and a new precursor compound of antigen of estrone(3-1)were studied in this thesis.It consists of the following three chapters.The first chapter briefly introduced the discovery,structure,bioactivity,biosynthetic pathway of estrone and summarized the main chemical synthetic strategies.In the second chapter,aiming at searching for a new synthetic strategy of estrone,a new synthetic strategy was designed from available 1,4-cyclohexanedione.The key intermediate(2-2)was obtained from available 1,4-cyclohexanedione via monoketalization,Robinson cyclization,Birch reduction and off-protecting group.Further more,we tried to rapidly construct the A-ring and D-ring in the steroid structure through the double Robinson cyclization reaction of(2-2).In the third chapter,a new precursor compound of holoantigen of estrone(3-1)was been synthesized from estrone(1)by reaction with adding protection group,oxidation,reduction,nucleophilic substitution,off-protecting group and hydrolysis.
