Rice is one of the most difficult to store grain.Due to the loss of rice hull protection,resulting in nutrients directly exposed.It is sensitive to temperature,humidity and gas composition.And easy to be affected by damp,heat,insect and mould,which affects the quality of rice.The storage of rice is a major problem in the field of grain storage.It is important contents of rice storage to safe storage,mildew prevention and quality deterioration.In this research,electron beams of 0k Gy,1k Gy,3k Gy,and 5kGy were used to irradiate rice.The acute oral toxicity test in rats and mice,genetic toxicity test(Ames test,bone marrow cell micronucleus test and mice sperm abnormality test)and 28 days oral toxicity test were conducted according to the Food Safety Assessment of genetic toxicity evaluation procedures and methods.Objective to comprehensively evaluate the edible safety of electron beam irradiated rice.The following conclusions are drawn in this study:(1)There were no death occurred in all the rats and mice during the observation of(1)There were no death occurred in all the rats and mice during the observation of acute oral toxicity test.And in the whole acute toxicity test,the animal skin,hair,eyes,mucous membrane and so on,as well as the anatomy of the organs,diet and did not find abnormal phenomenon.The results showed that the oral LD50 of female and male mice irradiated by 1,3 and 5kGy3 doses were higher than 10g/kg.BW.According to the LD50 dose grading standard,the samples are all non-toxic.(2)Genetic toxicity test of rice irradiated by electron beam The conclusion was drawn according to the results of bacterial reverse mutation test(Ames test): With the increase of dose,the number of mutant colonies changed little and did not cause a large number of cell recovery mutation.It is indicated that the subject is not mutagenic under the experimental conditions.Electron beam irradiation rice mouse bone marrow polychromatic erythrocyte micronucleus test was negative.The test can not cause the increase of mammalian polychromatic erythrocytes with micronucleus cell rate,no mutagenic effect.We can draw a conclusion according to the results of mice sperm abnormality test.The mice sperm abnormality test was negative in this experimental dose range,and no significant damage to male mouse germ cells was observed after electron beam irradiation.Three genetic test(Ames test,bone marrow cell micronucleus test and mice sperm abnormality test)the results were negative,the results showed that electron beam irradiation of rice showed no genetic toxicity in this test condition.(3)28 days oral toxicity of electron beam irradiation rice rat test No animal death occurred in the experiment.The general condition,weight change,food utilization rate,hematology and biochemistry indexes and test results were all in the normal range.Gross anatomy and pathological examination showed no obvious pathological changes and suspicious pathological changes.The results showed that electron beam irradiation had no obvious toxic effect on rat organs.To sum up,the electron beam irradiated rice has the edible safety when the irradiation dose is below 5kGy.
