In this experiment,peanut as raw material,the extraction of peanut in nicotinic acid by ultrasonic assisted by Box-Behnken response surface design method to establish the influencing factors of two regression model,and analysis of the optimum extraction conditions of ultrasonic assisted extraction of peanut nicotinic acid obtained by response surface data.Furthermore,the effect of lipid lowering and the antagonism of folic acid in peanut were studied,and the preparation process and formulation of nicotinic acid effervescent tablets were discussed.The main conclusions of this study are as follows:(1)On the basis of single factor experiment,65% ethanol solution was selected as extractant.The Box-Behnken of ultrasonic assisted extraction technology was optimized by response surface design method,the optimum conditions of ultrasonic assisted extraction of peanut by nicotinic acid: liquid ratio(mL/g)of 25.46:1,ultrasonic power 407.10 W,ultrasonic time was 16.98 min.According to the above conditions,the best process is the liquid to material ratio(mL/g)of 25:1,ultrasonic power of 405 W,ultrasonic time is 17 min,the yield of peanut niacin can reach 14.50(± 0.36)mg/100g(n = 3).(2)The study on the effect of the extract of peanut niacin on blood lipid lowering,the results showed that the indexes of the blood in the mice were significantly different from those of the control group.Total cholesterol decreased from 2.07nmol/L to 1.37nmol/L,the decline in the amount of up to 33%,the difference was significant;triglyceride decreased from 0.68nmol/L to 0.48nmol/L,the decline in the amount of up to 29%,the difference was significant;high density lipoprotein increased from 0.32nmol/L to 0.40 nmol/L,increased by 25%,the difference was significant;and mouse atherosclerosis index(AI)fell from 5.47 to 2.43 55%,the decline significantly.(3)Study on interaction of nicotinic acid and folic acid,niacin and folic acid intake at the same time,nicotinic acid content in mice blood average increased from 13.84nmol/L to 23.65nmol/L,increased by 70.9%,the difference was significant;the average content of folic acid increased from 3.70nmol/L to 8.82nmol/L,increased by 138.4%,significant difference.Compared with the normal control group,the values of the two substances were close to that of the intervention group respectively,indicating that the two substances did not antagonize each other at the same time.When the vitamin C and niacin folic acid together,the blood folic acid in mice increased from 3.70nmol/L to 7.26nmol/L,increased by 96.2%,the difference was significant.However,folic acid content was still significantly lower than the normal group and folic acid intervention group,indicating that in the presence of vitamin C,and folic acid antagonism.(4)To extract peanut nicotinic acid as raw materials,the preparation of nicotinic acid and vitamin C effervescent tablets.Determination of effervescent tablets of raw materials and processing through the query data,the best preparation process was determined by orthogonal experiment.When citric acid was added to 40 g,the content of NaHCO3 was 40 g,adding amount of binder is 8g,the effervescent tablet disintegration time shortest.(5)This experiment using the extracted peanut peanut meal nicotinic acid extraction of peanut protein isolate,by single factor of various factors on Extraction of peanut protein isolate in the process of extraction rate.Select and choose the 4 most important factors for test analysis of orthogonal test,the optimum conditions were obtained peanut protein isolate as the solid-liquid ratio is 12:1,alkali soluble pH9.5,soluble pH5.0,alkali solution temperature is 45 degrees centigrade,the alkali soluble 50 min.Under this condition,the extraction rate of peanut protein isolate was 82.56±1.26%(n=5).Peanut protein content 89.18%.
